Produção, purificação parcial e caracterização de xilanase produzida por fungo filamentoso

Abstract

Os fungos filamentosos são microrganismos muito eficientes na produção de xilanases, enzimas que apresentam diversas aplicações biotecnológicas, tais como, branqueamento do papel, aditivos em rações animais, panificação, extração e clarificação de sucos, entre outras. Este trabalho teve como objetivo selecionar e otimizar as condições de cultivo do microrganismo de maior potencial para a produção de xilanase, caracterizando a mesma bioquímicamente, e purificar parcialmente a de interesse. Onze fungos isolados em áreas de Cerrado, no estado do Mato do Grosso Sul, com crescimento a 30 ºC, foram reativados para ensaio de produção da xilanase. A maioria dos isolados produziram xilanase por cultivo em estado sólido, inicialmente, utilizando o farelo de trigo como substrato, se destacando três espécies identificadas como IPS8.2, IPS14.3F e IPS 9.3. O fungo IPS8.2 foi selecionado para estudo posteriores e identificado como Gongronella butleri com base sequenciamento de rDNA por ITSs. Além do cultivo inicial com farelo de trigo, esta espécie foi submetida ao efeito de diferentes fontes de carbono com outros substratos, tais como: casca de maracujá, serragem, papelão, casca de soja, sabugo triturado, bagaço de cana-de-açúcar, braquiária, sabugo de milho e xilana, nos cultivos em estado sólido (CES) e submerso (Csm). Os resultados obtidos nessa etapa não sobrepuseram à utilização inicial, em farelo de trigo em CES. Partindo desses resultados buscou-se verificar o efeito da concentração de sais, umidade e tempo no cultivo em estado sólido. Entre as condições testadas, Gongronella butleri IPS8.2 obteve sua melhor produção de xilanase em 72 horas de cultivo, com água destilada e 70% de umidade. A solução enzimática bruta, foi submetida à purificação parcial por troca iônica em Hitrap Q-Sepharose FF. As atividades da xilanase bruta e parcialmente purificada foram favorecidas à 45 ºC e pH 5,5. A enzima parcialmente purificada apresentou uma estabilidade de temperatura e uma faixa de estabilidade de pH inferiores à amostra bruta. A maioria dos íons utilizados não inibiu a xilanase bruta e à parcialmente purificada. Para ambas as condições testadas, o íon Ag+ foi o que proporcionou maior diminuição na atividade da enzima em estudo.

Filamentous fungi are very efficient microorganisms in the production of xylanases, enzymes that present several biotechnological technologies, such as paper bleaching, additives in animal feed, breads, extraction and clarification of juices, among others. The goal of this work was to select and optimize the culture conditions of the microorganism with the greatest potential for the production of xylanase, characterizing the same biochemically, and to partially purify the one of interest. Eleven fungi, with growth at 30ºC, isolated in Cerrado areas, in the state of Mato Grosso do Sul, were reactivated for the production of the xylanase. The majority of isolates produced xylanase by solid-state cultivation, initially using wheat bran as substrate, highlighting three species identified as IPS8.2, IPS14.3F and IPS 9.3. The fungus IPS8.2 was selected for further study and identified as Gongronella butleri, based on rDNA sequencing by ITSs. In addition to the initial cultivation with wheat bran, this species was submitted to the effect of different sources of carbon with other subtracts, such as passion fruit peel, sawdust, cardboard, soybean hull, crushed cob, sugar cane bagasse, corn cob and xylan, in the solid state cultivation (SSC) and submerged culivation (SC). The obtained results at this stage did not overlap with the initial use of wheat bran at SSC. Based on these results, was tried to verify the effect of the concentration of salts, humidity and time in the solid state cultivation. Among the tested conditions, Gongronella butleri IPS8.2 obtained its best xylanase production at 72 hours of culture with distilled water and 70% of humidity. The crude enzymatic solution, was subjected to partial purification by ion exchange in Hitrap Q-Sepharose FF. The activities of crude and partially purified xylanase were favored at 45 °C and pH 5.5. The partially purified enzyme had a temperature stability and pH stability range lower than the crude sample. The majority of the ions used didn‟t inhibit the crude and partially purified xylanase. For both conditions tested, the Ag+ ion provided the greatest decrease in the activity of the enzyme under study
