Crioconservación de semillas de Teca (Tectona grandis).

Abstract

La teca (Tectona grandis L.f) es un árbol tropical con gran demanda por la alta calidad de su madera y rápido crecimiento. Los programas de mejoramiento genético para esta especie han dado como resultado semillas de mejor calidad y, al mismo tiempo, el incremento en la calidad de las plantaciones. Con el objetivo de conservar la diversidad genética y garantizar la materia prima para los programas de mejoramiento y reproduc ción futura, las semillas de teca son mantenidas en bancos de semillas convencionales bajo con diciones de temperatura que oscilan entre 4º y –20ºC. Sin embargo, hay otras modalidades que pueden emplearse para complementar la conser vación de este valioso germoplasma. La criocon servación o almacenamiento en nitrógeno liquido (NL, -196ºC) presenta importantes ventajas sobre otras técnicas de conservación, como la posibili dad de almacenamiento por tiempo indefinido y en condiciones de alta estabilidad genética. En este estudio se evaluó la sobrevivencia y regene ración de plantas después del congelamiento de las semilas en nitrógeno líquido (NL), con el uso de la técnica de desecación y congelamiento rápi do. La metodología fue evaluada tanto en semi llas aisladas del endocarpo (semillas) como en semillas rodeadas por el endocarpo (semillas con endocarpo), que permite observar tasas de ger minación de 84% y 70% respectivamente, a los 28 días en cultivo, después de la descongelación.

Cryopreservation of teak (Tectona grandis L.f) seeds. Teak (Tectona grandis L.f) is a tropical tree of commercial value due to the high demand of its high-quality wood and rapid growth. Genetic improvement programs for this species have resulted in seeds of better quality and, at the same time, improvement in the quality of plantations. To preserve genetic diversity and guarantee the raw material for improvement programs and for future reproduction, seeds are kept under conventional seed banks conditions with temperature ranging between 4 and –20ºC. However, there are other means to conserve this valuable germplasm. Cryopreservation is the storage of plant material in liquid nitrogen (NL, -196ºC) and its major advantage is the conservation for indefinite periods of time, under high genetic stability conditions. Survival and regeneration of plants after seed freezing in liquid nitrogen (LN) were evaluated in this work, using the dessiccation and rapid freezing technique. The methodology was tested both, in seeds isolated from endocarps (seeds) asin seeds inside endocarps (seeds with endocarps), with germination rates, after thawing and 28 days in culture, of 84% and 70% respectively.
