Abstract

O presente trabalho teve como objetivo o desenvolvimento, implementação e tentativa de validação de uma técnica de cultivo e quantificação de esporos de Clostridium perfringens (estirpe ATCC 13124 / WDCM 00007), uma bactéria anaeróbia obrigatória frequentemente associada a surtos de origem alimentar. Devido à sua capacidade de formar esporos resistentes a condições adversas, torna-se fundamental dispor de métodos analíticos robustos e validados para a sua deteção em matrizes alimentares. Este estudo enquadra-se no contexto da garantia da qualidade analítica em laboratórios de microbiologia alimentar, tendo por base os requisitos da norma ISO/IEC 17025:2017, que estabelece critérios de competência para laboratórios de ensaio.A metodologia foi baseada na aplicação da norma ISO 15213-1:2023, direcionada à enumeração de bactérias sulfito redutoras, como C. perfringens, em meio TSC sob condições anaeróbias. A norma foi adaptada com o intuito de desenvolver um método quantitativo específico para esporos desta espécie. Esta adaptação, contudo, configurou-se como uma tentativa de validação de um novo método, exigindo uma abordagem detalhada conforme os princípios definidos nas normas ISO 16140-2:2016/Amd 1:2024 e ISO 16140-6:2019, direcionadas à validação de métodos microbiológicos quantitativos.Ao longo do estágio, realizaram-se ensaios para otimização de diversos parâmetros críticos, incluindo condições de esporulação, tempo e temperatura de tratamento térmico, eficácia da anaerobiose, volumes de inoculação, tempos de incubação e estado nutricional dos meios utilizados. Testaram-se ainda adaptações ao protocolo, como a extensão do tempo de banho-maria e variações de condições térmicas e nutricionais. No entanto, os ensaios demonstraram que o processo de esporulação e germinação de C. perfringens é altamente sensível a pequenas variações experimentais, não se tendo obtido uma germinação suficientemente consistente para suportar a validação experimental do método.Apesar disso, o trabalho apresenta uma abordagem pormenorizada sobre os parâmetros de desempenho a considerar numa eventual validação: exatidão, precisão (repetibilidade, reprodutibilidade, precisão intermédia), linearidade, limites de deteção e quantificação, robustez, erro sistemático e incerteza de medição. Estes aspetos foram explorados teoricamente com base nas normas ISO 19036:2019, ISO 7218:2024 e ISO 3534-2:2006, estabelecendo os procedimentos a seguir e os critérios de aceitação internacionalmente reconhecidos.Apesar de não ter sido possível validar experimentalmente a técnica, o estudo representa uma contribuição significativa para a compreensão dos desafios técnicos e normativos relacionados à quantificação de esporos de C. perfringens. Este trabalho representa uma base sólida para futuras investigações, salientando o compromisso com as boas práticas laboratoriais e os sistemas de gestão da qualidade microbiológica.

The aim of this study was to develop, implement and attempt to validate a technique for cultivating and quantifying spores of Clostridium perfringens (strain ATCC 13124 / WDCM 00007), an obligate anaerobic bacterium frequently associated with foodborne outbreaks. Due to its ability to form spores that are resistant to adverse conditions, it is essential to have robust and validated analytical methods for its detection in food matrices. This study falls within the context of analytical quality assurance in food microbiology laboratories, based on the requirements of the ISO/IEC 17025:2017 standard, which establishes competence criteria for testing laboratories.The methodology was based on the application of the ISO 15213-1:2023 standard for the enumeration of sulphite-reducing bacteria, such as C. perfringens, in TSC medium under anaerobic conditions. The standard was adapted in order to develop a specific quantitative method for spores of this species. This adaptation, however, was an attempt to validate a new method, requiring a detailed approach in accordance with the principles defined in ISO 16140-2:2016/Amd 1:2024 and ISO 16140-6:2019, aimed at validating quantitative microbiological methods.Throughout the internship, tests were carried out to optimise various critical parameters, including sporulation conditions, heat treatment time and temperature, the effectiveness of anaerobiosis, inoculation volumes, incubation times and the nutritional status of the media used. Adaptations to the protocol were also tested, such as extending the water bath time and variations in thermal and nutritional conditions. However, the tests showed that the process of sporulation and germination of C. perfringens is highly sensitive to small experimental variations, and germination was not sufficiently consistent to support experimental validation of the method.Despite this, the work presents a detailed approach to the performance parameters to be considered in a possible validation: accuracy, precision (repeatability, reproducibility, intermediate precision), linearity, detection and quantification limits, robustness, systematic error and measurement uncertainty. These aspects were explored theoretically on the basis of the ISO 19036:2019, ISO 7218:2024 and ISO 3534-2:2006 standards, establishing the procedures to be followed and the internationally recognised acceptance criteria.Although it was not possible to experimentally validate the technique, the study represents a significant contribution to understanding the technical and regulatory challenges related to the quantification of C. perfringens spores. This work represents a solid basis for future research, underlining the commitment to good laboratory practices and microbiological quality management systems.
