Abstract

Fastq files and sample info list from RNAseq experiment (November 2022) on tomato moneymaker wild-type (C32) and a mutants line for a TF involved in glandular trichome development and specialised metabolism (MS5). The mutant line has been transformed with (A) a construct for the \(\beta\)-estradiol inducible expression of the TF (to obtain complementation) or (E) with the empty construct as control. Plant material: sterilized and sanitized seeds were germinated in 1/2 agar MS medium. One week after germination, plants were transferred on either inducing medium with \(\beta\)-estradiol or with mock treatment. Then, 1 cm long root-tips were collected in triplo and pooled, at 0h, 12h and 24h after treatment, respectively. Root material was immediately snap-frozen in N2, homogenized with tissue lyzer and stored at -80 C. RNA extraction: Macherey-Nagel NucleoSpin RNA XS Kit (50) (Cat No./ID: 740902.50) for RNA isolation and purification according to plant protocol from producer. Library prep: poly-A enrichment from 500 ng RNA using the NEBNext Poly(A) mRNA Magnetic Isolation Module, then NEBNext Ultra II Directional RNA Library Prep and Multiplex Oligos kits for Illumina (Unique Dual Index Primer Pairs) (New England BioLabs). Libraries were clustered and sequenced (75 bp) on a NextSeq 550 Sequencing System (Illumina) using a NextSeq 500/550 High Output Kit v2.5 (75 Cycles) (Illumina).
