Abstract

Zusammenfassung Auf der Grundlage vorangegangener Modelluntersuchungen wurde versucht, zur Klarung der histochemischen Erfassungsmoglichkeit der Enzyme des Xanthindehydrogenase-/Xanthinoxidase-Systems beizutragen. Nach den Ergebnissen last sich an nativen Schnitten der Leber, der Niere und des Duodenums erwachsener mannlicher Wistarratten mit NBT und Hypoxanthin kein positiver Reaktionsausfall erzielen. Ohne Wirkung bleiben auch die Effektoren PMS sowie Triton X 100 und Tween 80 . Nach kurzzeitiger (10 min) Glutaraldehyd-Fixierung der Schnitte zeigt sich in den genannten Organen, besonders stark im Duodenum, ein positiver Reaktionsausfall, der unter dem Einflus von PMS bzw. der Defcergentien Triton X 100 und Tween 80 eine deutliche Verstarkung erfahrt. Die Membrantechnik bringt methodisch keine Vorteile beim histochemischen Nachweis der Enzyme des XDH-XOD-Systems. Die Spezifitat des erzielten Reaktionsausfalls wird unter den Aspekten der aus biochemischen Untersuchungen bekannten Komplexitat des XDH-/XOD-Systems kritisch besprochen.
