Dynamics of Nucleic Acids and Total Proteins in the Thymus and the Bursa of Fabricius of Chickens, under the Action of Cortisol

Abstract

Summary 5 day old chickens of the Studler strain were injected i. m. with a single dose of 3.5 mg. cortisol per 100 g. body weight. They were sacrificed at 8 and 24 hrs, and at 3, 6, 14, and 28 days. Involution phenomena were observed both in the thymus and in the bursa of Fabricius, with maximal intensity on the 6th day after hormone administration. The weight of both glands, as well as their concentrations of total nucleic acids, RNA and DNA, significantly decrease as compared to the controls. Later, the levels return gradually to normal, which reflects regeneration of the thymus and bursa. By the 28th day the values were normal. Total proteins show an increase during the first 24 hrs after exogenous cortisol loading. The return to normal of this parameter is faster in the thymus than in the bursa. Zusammenfassung Dynamik der Nukleinsauren und des Eiweisgehaltes im Thymus und in der Bursa fabricii der Huhnchen unter dem Einflus von Cortisol 5 Tage alten Kuken des Studler Stammes wurde eine einmalige intramuskulare Injektion von 3,5 mg Cortisol pro 100 g Korpergewicht verabreicht. Nach 8 bzw. 24 Stunden sowie nach 3, 6, 14 und 28 Tagen wurden die Tiere getotet. Sowohl am Thymus als auch an der Bursa fabricii wurden Involutionserscheinungen beobachtet, welche am 6. Tag nach der Hormon-applikation ihre groste Intensitat erreichten. Das Gewicht beider Drusen sowie die Totalkonzentration der Nukleinsauren (RNS und DNS) waren im Vergleich zu den Kontrolltieren signifikant vermindert. Die Parameter normalisierten sich sukzessiv, was auf eine Regeneration des Thymus und der Bursa hinweist. Nach 28 Tagen waren beide Werte wieder normal. Wahrend der ersten 24 Stunden nach der Cortisol-Applikation erhohte sich der Total-proteingehalt. Die Ruckkehr dieses Parameters zum Normalwert erfolgte im Thymus rascher als in der Bursa. On a sacrifie les animaux apres 8 et 24 heures ainsi qu'apres 3, 6, 14 et 28 jours. On a observe des symptomes d'involution aussi bien du thymus que de la bourse de Fabricius qui ont atteint leur plus grande intensite au 6eme jour apres l'application d'hormone. Le poids et la concentration totale des acides nucleiques (ARN et ADN) des deux glandes etaient diminues de facon significative par rapport aux animaux de controle. Les parametres se normaliserent successivement, ce qui tend a demontrer une regeneration du thymus et de la bourse de Fabricius. Les valeurs etaient a nouveau normales apres 28 jours. Le total des proteines s'est eleve dans les 24 premieres heures apres l'application de cortisol. Le retour a la normale de ce parametre s'est effectue plus rapidement pour le thymus que pour la bourse de Fabricius. Resumen Dinamica de los acidos nucleicos y de las proteinas totales en el timo y en la bolsa de Fabricio de pollitas bajo la accion de cortisol Se aplico a pollitas de 5 dias de edad de la estirpe Studler una inyeccion intramuscular unica de 3,5 mg de cortisol por 100 g de peso corporal. Tras 8 resp. 24 horas, asi como despues de 3, 6, 14 y 28 dias se mataron los animales. Tanto en el timo como en la bolsa de Fabricio se observaron fenomenos de involucion, los cuales alcanzaron su intensidad maxima el 60 dia tras la aplicacion hormonal. El peso de ambas glandulas, asi como la concentracion total de los acidos nucleicos (ARN y ADN) se hallaban disminuidos de modo significante en comparacion con los animales testigos. Los parametros se normalizaban gradualmente, lo cual refleja la regeneracion del timo y de la bolsa de Fabricio. Tras 28 dias estaban normalizados ambos valores. Durante las 24 horas primeras despues de aplicar el cortisol aumentaba el contenido en prcteinas totales. La vuelta de estos parametros a los valores normales se efectuaba de forma mas rapida en el timo que en la bolsa de Fabricio.
