Lipoik asit: Evrensel antioksidan

Abstract

Alpha-lipoic acid is a natural substance found in a variety of foods and also sythesized in the human body. It prevents free radical damage and it is unique among anti-oksidants because of its solubility in both oil and water. In humans, lipoic acid is part of various 2-oxo-acid dehydrogenases that play a key role in energy production. There are two forms of alpha-lipoic acid:oxidized and reduced lipoic acid. The reduced form is also called as dihydrolipoic acid. Dihydrolipoic acid is biologically more active than the reduced lipoic acid. Lipoic acid is sythesized in mitochondria from oktanoic acid and a sulfur source. When given orally, more than 93 percent of it is absorbed by the intestine and its first-pass effect by the liver is 20 to 30 percent. The primary metabolic pathway of alpha-lipoic acid in humans is Smethylation and β-oxidation. Lipoic acid acts as a carrier and transports two electrons. It participates in alpha-ketoglutarate dehydrogenase and pyruvate dehydrogenase enzyme complexes. Lipoic acid acts as a coenzyme in oxidative decarboxylation of pyruvate. Lipoic acid has been shown to regenarate Vitamin C and E which prevents free radical damage.

α-Lipoik asidin insanlardaki primer metabolik yolu S-metilasyon β-oksidasyondur. Lipoik asit açil taşıyıcı olup iki elektron taşımakla görevlidir. α- ketoglutarat dehidrogenaz ile pirüvat dehidrogenaz enzim kompleksleri içinde bulunur. Lipoik asit pirüvatın oksidatif dekarboksilasyonunda koenzim olarak görev yapmaktadır. Lipoik asidin serbest radikal hasarını uzaklaştıran vitamin E ve vitamin C’ yi rejenere ettiği gösterilmiştir.
