Confronto di marcatori molecolari per l'identificazione simultanea di Vibrio cholerae, V. parahaemolyticus e V. vulnificus tramite multiplex-PCR.

Abstract

Tra i batteri presenti in ambiente acquatico ed associati a molluschi bivalvi, le specie potenzialmente patogene per l'uomo Vibrio cholerae, V. parahaemolyticus e V. vulnificus rappresentano una problematica emergente da un punto di vista igienico-sanitario. L'inadeguatezza delle tecniche attualmente utilizzate presso i laboratori diagnostici per l'identificazione di tali batteri rende necessario l'utilizzo di tecniche molecolari basate sulla polimerase chain reaction (PCR) e l'individuazione di marcatori molecolari adeguati per l'identificazione a livello di specie. Due geni per ciascuna specie sono stati testati in 4 ceppi di riferimento e 136 ceppi di campo isolati da molluschi e acqua marina, precedentemente identificati su base biochimica come V. cholerae, V. parahaemolyticus, V. vulnificus e V. alginolyticus. Gli esiti delle singole PCR sono stati concordanti per V. cholerae. Per quanto riguarda V. vulnificus e V. parahaemolyticus uno dei due marcatori è risultato inadeguato, nel primo caso per la presenza di bande aspecifiche, nel secondo poiché un gene risulta positivo anche nella specie V. alginolyticus. Sulla base dei risultati ottenuti sono stati scelti come marcatori molecolari toxR per V. cholerae, toxR per V. parahaemolyticus e vvhA per V. vulnificus per la messa a punto di un protocollo di multiplex-PCR per l'identificazione simultanea dei tre batteri potenzialmente patogeni.
