Abstract

PAMUKKALE ÜN VERS TESFEN B L MLER ENST TÜSÜTEZ ÖZET B LD R FORMUB YOLOJ ANAB L M DALIDOMATESTE PATATES Y V RÜSÜ (PVY) DAYANIKLILIĞININ GENET ĞEminur BARUTÇUÖZETPatates Y virüsü (PVY), önemli besin kaynaklarından olan domates bitkisi içinönemli bir patojendir. Bu patojenin saldırısından bitkiyi korumak için bu çalışmada, 50farklı yabani tür ve kültür domates türlerinde PVY'ye karşı dayanıklılık kaynaklarıaraştırılmıştır. Yabani ve kültür türleri ve bu hatların çaprazlarından elde edilen F1 ve F2hatları PVY ile mekanik olarak inokule edilmiştir. nokule edilen bitkiler, iki haftalık ve dörthaftalık dönemlerde DAS-ELISA (Double Antibody Sandwich Enzyme-linkedimmunosorbent assay) ile test edilmiştir. Yabani domates türlerinde değişik şekillerdedayanıklılık tespit edilmiştir. Lycopersicon chilense ve L. pennellii ve L. hirsutum PI247087bitkisinde PVY'ye karşı immünite olduğu gözlenmiştir. L. peruvianum, L. pimpinellifolium,L. glandulosum, L. chmielewskii bitkilerinde PVY'ye karşı dayanıklılık olduğu tespitedilmiştir. L. hirsutum ve L. chmielewskii F2 populasyonlarının fenotipik analizlerine göredayanıklılığın resesif tek genle kontrol edildiği varsayılmaktadır. Sonuçlar Ki-kareanalizinde belirlendiği gibi 1:3 açılım oranına uymaktadır.

PAMUKKALE ÜN VERS TESFEN B L MLER ENST TÜSÜTEZ ABSTRACT B LD R FORMUGRADUATE PROGRAM IN BIOLOGYTHE GENETICS OF POTATO VIRUS Y (PVY) RESISTANCE IN TOMATOEminur BARUTÇUABSTRACTPotato virus Y (PVY) is an important pathogen for tomato which is an important crop for thehuman diet. In this study, resistance against PVY was surveyed in 50 different wild and cultivatedtomato types to identify sources for protection against this disease. Wild type tomatoes andcultivars and their F1 hybrids and F2 progenies were mechanically inoculated with PVY. Inoculatedplants were evaluated by DAS-ELISA (Double Antibody Sandwich Enzyme-linked immunosorbentassay) after two and four weeks. Different types of resistance were observed in wild types.Immunity to PVY was observed in Lycopersicon chilense, L. hirsutum PI247087 and L. pennelliiplants. Resistance to PVY was observed in L. peruvianum, L. pimpinellifolium, L. glandulosum, L.chmielewskii plants. According to phenotypic analysis of L. hirsutum and L. chmielewskii F2populations, it was hypothesized that resistance was controlling by a single recessive gene becauseresults fit a 1:3 segregation ratio as determined by Chi-square goodness-of-fit analysis.
