Abstract

In this study,23 previously identified Yarrowia lipolytica strains were evaluated forthe organic acid (sitrikasit, izositrik asit) production capacities and two strains (Yarrowia lipolytica TEMYL 3 ve Yarrowia lipolytica TEMYL 20) which have the highest organicacidproducing capable were selectedfor further experiments in the research.Productionwascarried out approximately 408 hours in a batch system and amounts of citric/isocitric produced by Y.lipolytica TEMYL3 and TEMYL20 strains were determined enzymatically.In study,glucose, glycerol and sunflower oil were used as substrates and the initial concentrationof carbon sources in the fermentation media wereadjusted as 50 and 100g/L.For the Y.lipolytica TEMYL3, in media containing 50g/L glucose, glycerol and sunflower oil, the highest citric acidproduction amounts were 18.24, 6.50, 22.30 g/L, respectively. Also,for the strain TEMYL 3, maximum citric acid production amounts were determined as 33.30,35.60 and 66.20g/L in medium containing 100 g/L glucose, glycerol and sunflower oil, respectively. For the Y.lipolytica TEMYL20, the highest citric acidproduction amounts were obtained as 17.36, 6.74 and 31.41 g/L in medium containing 50g/L glucose, glycerol and sunflower oil, respectively. Also, for the strain TEMYL 20, in media containing 100 g/L glucose, glycerol and sunflower oil ,maximum citric acid production amounts were 36.30,37.50 and 53.76 g/L, respectively. Additionally, for the Y.lipolytica TEMYL20,maximum isocitric acid production amount was obtained as 53.70 g/L in medium containing 100g/L sunflower oil and the lowest isocitric acid production (1.09 g/L) was obtained in medium containing 50g/L glycerol.Keywords: Citric acid, isocitric acid, yeast, Yarrowia lipolytica

Bu araştırmada, başka bir çalışmada tanısı yapılmış olan 23 adet Yarrowia lipolytica strainlerinin organik asit üretim kapasiteleri değerlendirilmiş ve aralarından en yüksek organik asit (sitrik asit, izositrik asit) üretme yeteneğine sahip olan iki strain (Yarrowia lipolytica TEMYL3 ve Yarrowia lipolytica TEMYL20) bu araştırma için seçilmiştir.Üretim kesikli bir sistemde yaklaşık 408 saat gerçekleştirilmiş olup, Y.lipolytica TEMYL3 ve TEMYL20 strainlerinin sitrik ve izositrik asit üretimleri enzimatik olarak saptanmıştır. Çalışmada, substrat olarak50 ve 100g/L başlangıç konsantrasyonunda glukoz, gliserol ve ayçiçek yağı kullanılmıştır.Y.lipolytica TEMYL 3 straini ilemaksimum sitrik asit konsantrasyonu; 50g/L glukoz, gliserol ve ayçiçek yağı içeren ortamlarda sırasıyla 18.24, 6.50, 22.30 g/L olarak elde edilmiştir.
