Abstract

The objective of this study was to evaluate the exogenous fibrolytic enzymes production by fungal fermentation and the application in different doses at ensiling of maize. The project was divided into two sub projects, the first was characterized by enzymes production and their stability in rumen conditions. Five samples were collected from different locations and 45 fungi were isolated and evaluated for xylanases and cellulases production. Among the micro-organism selected as the best producer was a Aspergillus niger, capable to produce significant quantities of interest enzymes in this study (7,04U/mL and 0,088U/mL of xylanases and cellulases, respectivamente, respectively). After, followed by a test optimization of cultive conditions the fungus and enzymatic activity seeking maximum production of the interest enzymes. Optimal conditions of pH and temperature to enzymes reaction in this study corresponded to 4.6 and 3.6 for xylanase and cellulase, respectively, with optimum temperature of 60 ° C for both enzymes. The highest xylanases production occurred at 96 and 168 hours by submerged fermentation with or without agitation, respectively. Higher cellulase production was observed in 120 hours of cultive in two growth conditions (submerged with or without agitation). Optimal concentration of spores in the medium to be inoculated was 1.5 x107 and 5x107 spores / ml for xylanases and cellulases, specifically. Stability test of the enzymes in the rumen showed a greater xylanase activity in animals that received the enzyme extract via cannula until 7 hours after application. The second experiment evaluated the application of different levels of enzymes at ensiling of maize on the fermentation characteristics, chemical composition and aerobic stability of corn silage. O presente trabalho avaliou a produção de enzimas fibroliticas exógenas obtidas a partir da fermentação fúngica e sua aplicação em diferentes doses na ensilagem de milho. Para isso, o projeto foi dividido em dois experimentos, sendo o primeiro caracterizado pela produção de enzimas e sua estabilidade em condições ruminais. Cinco amostras de diferentes locais foram coletadas e 45 fungos foram isolados e avaliados quanto a produção de xilanases e celulases. O micro-organismo selecionado como o melhor produtor foi um Aspergillus niger, capaz de produzir significativas quantidades das enzimas de interesse neste estudo (7,04 e 0,09 U/mL de xilanases e celulases, respectivamente). Após, seguiu-se um ensaio de otimização das condições de cultivo do fungo e avaliação da atividade enzimática buscando a máxima produção das enzimas de interesse. As condições ótimas de pH e temperatura de reação das enzimas em estudo corresponderam a 4,6 e 3,6 para as xilanases e celulases, respectivamente, com temperatura ótima de 60ºC em ambas as enzimas. A maior produção de xilanases ocorreu com 96 e 168 horas na fermentação submersa com ou sem agitação, respectivamente. Na produção de celulases os maiores níveis enzimáticos foram observados com 120 horas de cultivo nas duas condições de crescimento (submersa com ou sem agitação). A concentração ideal de esporos a ser inoculado no meio foi de 1,5x107 e 5x107 esporos/mL nas xilanases e celulases, especificamente. O teste de estabilidade das enzimas no rúmen mostrou maior atividade de xilanases nos animais que receberam o extrato enzimático via cânula até 7 horas após a aplicação.
