Uso de sulfato de cálcio como redutor de estresse no transporte de matrinxãs (Brycon cephalus)

Abstract

The calcium sulfate or agricultural gypsum is used in aquaculture to reduce the stress responses during transportation, and the aim this study was to test its efficiency in 4h transport of matrinxã juveniles. After 48h in 500L aquaria, fasted fish (200.21g ± 51.12g) were transported for 4h in plastic bags (125g L-1) (five bags per treatment). One bag was sampled 15 min after package and the other were transported in water containing different CaSO4 concentrations: 0, 75, 150, 300 mg L- 1. Water samples of each treatment were evaluated: dissolved oxygen, pH, ammonia, hardness, alkalinity and calcium ions. Blood was collected before transport (initial value), after packing, after transport and 24 and 96 h later (recovery). Cortisol, chloride, sodium, potassium, calcium were determined in serum and hemoglobin. Red and white blood cells count, mean corpuscular volume, hematocrit, plasma glucose and osmolality were determined in eight fish (replicates) (2 fish per aquaria) before transport and recovery. Data were analyzed by ANOVA and means compared by Tukey test. Cortisol increased after package (control p<0,05; others p<0,01), but decrease after transport in fish transported with calcium sulfate. Glucose increased after transport in fish of all treatments and during recovery it decreased to values lower than those registered before transportation. After package the chloride levels showed tendency to decrease in treated fish, which elevating at arrival in control and in the two lowest concentration groups. In fish transported in 300 mg L-1, the chloride values were maintained similar to the initial levels. Sodium showed similar profile. Potassium presented tendency to decrease inversely to the concentration of calcium in the water and returned to initial values 24 h after. The serum calcium did not differ among treatments. O sulfato de cálcio (CaSO4) ou gesso agrícola é utilizado na piscicultura para minimizar as respostas de estresse durante o transporte. O objetivo deste estudo foi testar sua eficiência no transporte de 4 horas de matrinxãs juvenis. Após 48 horas de jejum, os peixes (200,21g ± 51,12g) foram embalados em sacos plásticos (125g L-1) (cinco sacos por tratamento), sendo um amostrado 15 minutos após a embalagem e 4 transportados em água contendo CaSO4 (0, 75, 150, 300 mg L-1). Foi retirada uma amostra de água das embalagens para avaliação do oxigênio dissolvido, pH, amônia, dureza, alcalinidade e íons cálcio. Amostras de sangue sem anticoagulante foram retiradas antes do transporte (nível inicial), após a embalagem, na chegada, 24 e 96 horas depois (recuperação). O soro foi separado para análise de cortisol, cloreto, sódio, potássio, cálcio e sangue heparinizado foi utilizado para avaliação do hematócrito, número de eritrócitos, número de leucócitos, volume corpuscular médio e hemoglobina no sangue total. No plasma foram dosados glicose e osmolalidade. Foram amostrados 8 peixes (repetições) (2 em cada caixa) antes do transporte e na recuperação, 8 em cada saco plástico após serem embalados e 8 (2 por saco plástico) após o transporte. Os dados foram submetidos à análise de variância pelo teste F, sendo as médias comparadas pelo teste de Tukey ao nível de 5% de probabilidade. Peixes de todos os tratamentos apresentaram aumento de cortisol após a embalagem (controle p<0,05; demais p<0,01) e na chegada na qual diminuiu nos peixes transportados com sulfato de cálcio. A glicemia aumentou após o transporte nos peixes de todos os tratamentos, sem diferença entre eles. Após 24 e 96 horas, diminuiu atingindo valores menores que os de antes do transporte.
