Abstract

The present research had as main objective the accomplishment studies that could elucidate some activities of muscular enzymes on proteins that determine meat tenderness. The Importance of the imunoistochemestry analyses is standed out, therefore specific related literature citations are rare on this subject about the modifications occured in the transformation process of muscle in meat at the first twenty four hours after slaughter of sheep. In the experimental assays, thirty crossbred male Suffolk sheep had been slaughtered using a humanitarian method. The slaughter method used was electronarcosis that influenciates in some meat quality factors. Longissimus dorsi fragment was excised from the carcass after 30 minutes after bleeding and maintained in a single individual identified plastic recipe at 2o a 4o C during 24 hours. After this, the fragments was intended and fixed in formalin solution (10%) to histological assay. pH an temperature was measured using a dry pHmeter equipment. Imunoistochemestry analysis were made by blocking endogenous peroxidasis using the proportion of 1:1 of hydrogen peroxide (20 v.) and methanol. After this procedures the tissue was incubated with primary antibodies to desmin and troponina C and T (1:100) in a 1%BSA solution at 4oC in a wet chamber about 18 hours. The tissue was colored using Diamino-Benzidin and colored again using Meyer hematoxilin to slide analysis. The carcasses pH and temperature analysis were measured at three moments and showed intensive decrease of values, mainly in initial twelve hours. After twenty four hours, the average of pH and temperature was presented in 5,51 and 4,58oC, respectively. Imunoistochemestry assessment had disclosed significant degradation of Desmin and Troponin T. This was not observed in the degradation of Troponin C. Statistics comparison of the three variables... O presente trabalho teve por principal objetivo a realização de estudos que elucidassem certas atividades das enzimas presentes nas células musculares, sobre as proteínas que determinam a maciez da carne. Ressalta-se a importância das análises imunoistoquímicas, pois foram raras as citações literárias específicas encontradas sobre este assunto, relacionadas às alterações ocorridas na carne de ovinos, no processo de transformação do músculo em carne nas primeiras vinte e quatro horas após o abate. Nos ensaios experimentais foram utilizados trinta ovinos machos mestiços da raça Suffolk que foram abatidos de forma humanitária. O método de insensibilização utilizado foi a eletronarcose que, sabidamente, influencia alguns fatores que irão determinar a qualidade da carne. Foi retirado um fragmento do músculo Longissimus dorsi de cada carcaça, aproximadamente 30 minutos após a sangria, de forma semelhante ao toilete de contusão da região lombar de cada carcaça,os quais foram acondicionados em recipientes plásticos individuais, com as devidas identificações e mantidos sob refrigeração (2o a 4o C), por 24 horas e depois recortados e fixados em solução de formalina tamponada a 10%, para a realização de cortes histológicos. A temperatura e pH das carcaças foram mensurados com a utilização de um potenciômetro portátil medidor de pH, com probe seca, que possui um termômetro em sua estrutura. As análises imunoistoquímicas foram realizadas a partir do bloqueio da peroxidase endógena: em partes iguais de metanol e água oxigenada 20 volumes, após, fez-se a incubação com anticorpos primários: desmina, troponina C e troponina T na diluição 1:100, que incubados em solução de BSA 1% a 4°C por 18 horas e em câmara úmida.
