Abstract

Alternatif akımlı (AC) elektromanyetik alanın ve doğru akımlı (DC) statik manyetikalanın mercimekte (Lens culinaris Medik.) mitotik indeks, kromozom davranışları vesitoplazmik kalsiyum dağılımı üzerindeki etkileri incelenmiştir. Mercimek tohumları petrikaplarında, laboratuar koşullarında, nemlendirilmiş filtre kağıtları üzerinde 3 günçimlendirildikten sonra manyetik alan uygulamalarına maruz bırakılmıştır. Uygulamadakontrol grubu, alternatif akımlı manyetik alan grubu ve doğru akımlı manyetik alan grubuolmak üzere üç grup kurulmuş ve her grup için 3, 6 ve 12 saatlik uygulamalar yapılmıştır.Alternatif akımlı elektromanyetik alanda 2,25 µWb ve 3,86 µWb olmak üzere iki, doğruakımlı statik manyetik alanda ise 300 G, 600 G ve 1000 G olmak üzere üç şiddetkullanılmıştır. Alternatif ve doğru akımın kullanıldığı uygulamalarda köklerde bazımorfolojik bozukluklar gözlenmiştir. Ezme preparatlarda mikronukleus oluşumu, ikinukleuslu hücreler, kromozom köprüleri, metafaz kromozomlarında aşırı kontraksiyon, geççekilen kromozomlar, iğ oriyentasyonu farklılığı gibi mitotik anormallikler saptanmıştır.Ayrıca nukleoplazmada vakuol oluşumu, sitoplazmada aşırı vakuolleşme, plazma zarında venukleus zarında bozukluklar da gözlenmiştir. Mitotik indeks şiddet ve süre artışına paralelolarak azalmış, 1000 G, 3 saatlik uygulamada ise sıfırlanmıştır. Manyetik alan uygulamasınınsitoplazmik kalsiyum dağılımına etkisini araştırmak üzere köklere Alizarin Red S boyamasıyapılmıştır. Şiddet ve süre artışına bağlı olarak sitoplazmik kalsiyumda artış tespit edilmiştir.Alternatif akımlı elektromanyetik alan ve doğru akımlı statik alan mercimekhücrelerinde, mitotik indeksi azaltmış, mitotik kromozomlarda ve sitoplazmadaanormalliklere neden olmuş, sitoplazmik kalsiyum dağılımını değiştirmiştir.

The effects of AC and DC magnetic fields on mitotic index, chromosomal behavior anddeposition of calcium ions in cytoplasm were investigated in lentil (Lens culinarisMedik.). Lentil seeds were germinated in petri dishes covered with fitler paper moistenedwith distilled water. 3 days after sowing,the rot tips were exposed to the AC and DC for 3,6 and 12 hours. Magnetic field by AC 2,25 µWb and 3.86 µWb, by DC 300G, 600 G ve1000G were used. Some morphological defects were observed on the roots exposed tomagnetic fields. Some mitotic abnormalities were also observed in treated roots such as,chromosome bridges, stickiness, lagging chromosomes, micronuclei, binucleate cell.Vacuolization is very conspicuous in the cytoplasm as well as in the nucleoplasm. Breakson the nuclear and plazma membranes were observed. Mitotic indices were decreased byincreasing time and dose and it fell zero at 1000 G. In order to identify calcium ionsdistribution in cytoplasm Alizarin Red S applied to the root tips. The root tips exposed tothe magnetic fields were more strongly stained than control roots depending on time anddose.In conclusion, In the root tips exposed to the AC and DC magnetic fields mitotic indexwas decreased, some abnormality on chromosomal bahavior and cytoplasm were observedand cytoplasmic deposits of calcium ions changed depending on time and dose.
