5. ÖZET Sıçan kalp ve karaciğer ve tavşan karaciğer dokularından toplam RNA elde edildi. Elde edilen RNA naylon membrana aktarılarak G3ot'ya özgü sentetik oligonükleotit prob ile hibritlemeye bırakıldı. Hibritleme sonucu sinyal gözlenmediği için aynı membran bu kez (CA) ^g probu ile etkileştirildi. Ayrıca kobay kalp ve karaciğer dokularından membran kesimleri elde edilerek S-142 hücre özütü kesimlerinde tüm ^-proteinlerine özgü antikor kullanılarak Western Blot analizi yapıldı. Karaciğer dokusundaki toplam G-protein miktarı kalp dokusundan fazla bulundu. 53
6 - SUMMARY Total RNA was obtained from rat heart, rat liver and rabbit liver and transferred to nylon membrane and allowed to hybridize with a synthetic oligonucleotide probe specific for G3a. No visible signal was detected; therefore, the membrane was hybridized with a (CA)xo probe. Membrane extracts were prepared from heart and liver tissues of Guinea-pigs. Western blot analysis using an antibody specific for Ga-subunits indicated that the total amount of G- proteins in liver was higher than that in heart.
