Abstract

Dr. Sevda Süzgeç SelçukFarmakognozi Anabilim DalıAmaç: Bu çalışmada Ferulago trachycarpa Boiss. bitkisinin toprak altı kısımlarından elde edilen n-hekzan, diklorometan ve metanol ekstrelerinin antimikrobiyal aktivitelerinin araştırılması ve aktif çıkan ekstreler üzerinde etken madde izolasyonu yapılması; izole edilen saf etken maddelerin antimikrobiyal aktivite açısından incelenerek ekstrelerdeki antimikrobiyal aktivite ile karşılaştırılması amaçlandı. Aynı zamanda F. trachycarpa bitkisinin toprak altı kısımlarından elde edilen su ve metanol ekstrelerinin antioksidan aktiviteleri çalışıldı. Gereç ve Yöntem: F. trachycarpa Boiss. bitkisinin toprak altı kısımları kullanıldı ve perkolasyon yöntemiyle n-hekzan, diklorometan ve metanol ekstreleri elde edildi. Elde edilen ekstrelerin çeşitli bakteri ve mantar suşlarına karşı mikrodilüsyon yöntemiyle antimikrobiyal aktiviteleri incelendi. Antimikrobiyal aktivite gösteren n-hekzan ve diklorometan ekstrelerinden etken maddelerin saflaştırılmasında çeşitli kromatografi yöntemleri (İTK, sütun, pİTK) kullanıldı. Saf olarak elde edilen maddelerin spektroskopik yöntemler (UV, IR, 1H-NMR, APT ve LC/MS) kullanarak yapıları aydınlatıldı. İzole edilen saf maddelerin antimikrobiyal aktiviteleri araştırıldı. Ayrıca F. trachycarpa bitkisinden perkolasyon yöntemiyle elde edilen su ve metanol ekstrelerinin antioksidan aktiviteleri DPPH, FRAP yöntemleri ile belirlendi ve toplam fenolik madde içeriği tespit edildi.Bulgular ve Sonuçlar: F. trachycarpa bitkisi n-hekzan ekstresinden kumarin bileşikleri olan krenulatin, suberosin, marmesin senesioat (prantşimgin) ile diklorometan ekstresinden ulopterol maddeleri izole edildi. Ferulago türlerinde, krenulatin ve ulopterol varlığı ilk defa bu çalışma ile gösterildi. Ulopterol dışındaki saf maddelerin hepsi, metisiline dirençli Staphylcoccus aureus bakteri suşuna karşı antimikrobiyal aktivite gösterirken; Candida albicans mantar suşuna karşı ise antifungal aktivite gösterdi. Su ekstresinin metanol ekstresine göre daha iyi antioksidan etki gösterdiği ve toplam fenolik madde miktarının daha yüksek olduğu tespit edildi. --------------------The Active Constituents Isolation from Ferulago trachycarpa Boiss. by Antimicrobial Activity GuidedTuğçe DikpınarThesis Advisor: Associate Prof. Dr. Sevda Süzgeç SelçukDepartment of PharmacognosyAim: The aim of this study was to make research about antimicrobial activities of n-hexane, dichloromethane and methanol extracts obtained from Ferulago trachycarpa Boiss., to do isolation on active substances and to investigate the antimicrobial activities of pure substances compare with extracts. At this time, the antioxidant activities of water and methanol extracts obtained from the rhizomes of F. trachycarpa were studied. Material and Methods: The rhizomes of F. trachycarpa Boiss. were used and obtained n-hexane, dichloromethane and methanol extracts by percolation method. The antimicrobial activities of obtaining extracts were investigated against bacteria and fungi strains using microdilution method. The various chromatografic methods (TLC, column, pTLC) were used to pure the active substances showing antimicrobial activity on n-hexane and dichloromethane extracts. The structures of substances obtained as pure identified by spectroscopic methods (UV, IR, 1H-NMR, APT and LC/MS). The antimicrobial activities of isolated pure substances were investigated. Besides, the antioxidant activities of water and methanol extracts obtained by percolation method were determined with DPPH and FRAP methods and total phenolic substance content were detected. Result and Conclusions: Coumarin derivatives; crenulatin, suberosin, marmesin senecioate (prantschimgin) were isolated from n-hexane extract and ulopterol was isolated from dichloromethane extract. Crenulatin and ulopterol have been first shown in Ferulago species with this study. Except ulopterol, while all pure substances showed antimicrobial activity against Staphylococcus aureus to methicillin resistant, they showed antifungal activity against Candida albicans fungi strains. Water extract was detected that showed the antioxidant effect better than methanol extract and had higher total phenolic substance content.
