Abstract

In this study, the genetic diversity of Tilia tomentosa Moench. (silver lime) species in Duzce province of Turkey was investigated by using RAPD markers. For this, a total of 28 leaf samples, including 25 samples from urban ecosystem and three samples from forest ecosystem were collected. DNA isolation of samples were performed by using `GeneJET Plant Genomic DNA Purification Mini Kit (Thermo Scientific USA)`. Eight primers were used as markers in RAPD method. A total of 53 different bands were obtained from 28 leaf samples and 48 of these bands were observed to be polymorphic. Thus, 90.6% of total number of bands was identified as polymorphic. The percentage of polymorphic loci (P) was found as 94.29%; number of alleles (Na) found as 1.94; effective number of alleles (Kimura ve Crow, 1964) (Ne) found as 1.60; Nei's genetic diversity (Nei, 1973) (h) found as 0.34 and Shannon's genetic diversity index (Lewontin, 1972) (I) found as 0.50. Moreover, UPGMA cluster and PCA analyses were performed in this study. Two main groups in UPGMA cluster analysis and three groups in PCA analysis were formed.

Bu çalışmada, Türkiye'nin Düzce ilindeki Tilia tomentosa Moench. (gümüşi ıhlamur) türünün genetik çeşitliliği RAPD işaretleyicileriyle araştırılmıştır. Bu amaçla 25'i şehir ve 3'ü orman ekosisteminden olmak üzere toplam 28 yaprak örneği toplandı. Örneklerin DNA izolasyonları `GeneJET Plant Genomic DNA Purification Mini Kit (Thermo Scientific USA)` kullanılarak yapıldı. RAPD yönteminde işaretleyici olarak 8 primer kullanıldı. 28 yaprak örneğinden, 53 farklı bant elde edildi ve bu batlardan 48'inin polimorfik olduğu görüldü. Böylece toplam bant sayısının %90.6'sının polimorfik olduğu tespit edildi. Polimorfik gen lokuslarının yüzdesi (P); %94.29, allel sayısı (Na); 1.94, etkili allel sayısı (Kimura ve Crow, 1964) (Ne); 1.60, Nei genetik çeşitlilik indeksi (Nei, 1973) (h); 0.34 ve Shannon genetik çeşitlilik indeksi (Lewontin, 1972) (I); 0.50 olarak hesaplandı. Çalışmada UPGMA kümeleme analizi ve PCA analizi yapıldı. UPGMA kümele analizinde iki ana grup, PCA analizinde ise üç grup oluştu.
