Identification of levansucrase gene in Halomonas sp.

Abstract

Fruktanlar, gelişmiş bitkilerin % 15' inin yani yaklaşık 40000 türün temel karbonhidrat kaynağını oluşturmakla birlikte, bakteri ile mantar türlerinin geniş bir kısmında bulunmaktadır.Fruktanların transfruktosilasyon reaksiyonuyla sentezi, sukroz molekülüne, hidrolize olmuş başka bir sukroz molekülünden gelen bir fruktosilin bağlanmasıyla başlar ve polimer zincirinin büyümesi, bağların tipi ve fruktan zincirinin dallanma özellikleri enzim ve organizmaya göre değişmektedir. Bakterilerde, çok fonksiyonlu bir adet enzim gerekmektedir. Bu enzimin adı; ürün bir fruktan tipi olan levan olduğunda levansukraz (EC 2.4.1.10), başka bir fruktan tipi olan inulin olduğunda ise inulosukraz (EC 2.4.1.9)' dır.Levansukraz, birçok mikroorganizma tarafından üretilmektedir ve bu enzimin katalitik aktivitesi ile sentezlenmiş levan polimerlerinin, kozmetik, gıda ve ilaç endüstrilerinde ve aynı zamanda hipokolesteromik ajan ve anti-tümör ajanı olarak bir çok potansiyel kullanım alanı da vardır. Levan yanında, bazı levansukraz enzimleri, potansiyel prebiyotik etkileri olan fruktooligosakkaritlerin üretimini de katalizlemektedir. Günümüzde, fruktooligosakkaritlerin endüstriyel üretimi esas olarak fungal levansukraz enzimine dayanmaktadır. Bundan dolayı, fruktooligosakkarit üretimini artıracak, bakteriyel yeni levansukraz gen kaynaklarının belirlenmesi, biyoteknolojik açıdan çok önemlidir. Halomonas sp., araştırma grubumuz tarafından ilk defa levan üreten bir mikroorganizma olarak tanımlanmıştır. Bu çalışmanın amacı ise, levansukraz enzimini kodlayan genin bakteri genomundaki lokasyonunun ve DNA dizisinin belirlenmesidir.Bu amaç doğrultusunda, Halomonas sp.' den genomik DNA izolasyonu ve primer tasarımından sonra, primerler kullanılarak optimize edilmiş PCR şartlarında gen bölgesinin seçici amplifikasyonu sağlanacaktır. Beklenen boyuttaki DNA bantları saflaştırılıp dizileri belirlenecektir.

Fructans constitute the primary carbohydrate reserve in 15% of higher plants comprising approximately 40000 species and appear in a wide range of bacterial and fungal species.The synthesis of fructans starts with a transfructosylation reaction in which a sucrose molecule plays the role of fructosyl donor and with a second molecule of sucrose as the initial acceptor of the fructosyl moiety. Following initiation, the extension, the type of linkage and the branching of the fructan chain varies according to the enzyme and organism. In bacteria, fructan biosynthesis is somewhat simpler than the synthesis in plants because only one multifunctional enzyme is involved. This enzyme is named levansucrase (EC 2.4.1.10) when the product is a fructan of the type levan or inulosucrase (EC 2.4.1.9) when the fructan is of the type inulin.Levansucrase is produced by various microorganisms and synthesizes a levan that has a variety of applications in the fields of cosmetics, foods and pharmaceuticals, as a hypo-cholesterolemic agent, and an anti-tumor agent. Hence the availability of levansucrase and its gene from bacteria as an alternate source of a fructooligosaccharide-producing enzyme might be of biotechnological interest. The use of levansucrase (EC 2.4.1.10) as a biocatalyst in levan synthesis has been the object of increasing interest in recent years. Halomonas sp. has been described as a levan producer microorganism for the first time by our research group. Consequently, the main objective of this thesis study was the identification of the gene coding for levansucrase activity by this microorganism.For this purpose, after purification genomic DNA from Halomonas sp. and design of primers, PCR conditions were optimized for the selective amplification of the regions enclosed by designed primers. The DNA bands at the expected size were purified and their DNA sequence was determined.
