`Conservation Biology? is the field of science studying the nature and the status of Earth's biodiversity with the purpose of developing strategies to protect species, their habitats and ecosystems from extinction. Conservation Genetics is the application of genetics to preserve species as dynamic entities capable of coping with environmental disturbances.The purpose of this dissertation is to contribute to the preservation of a endemic subspecies of Achilea multifida from extinction by using approaches of both conservation biology and genetics. Approximately 50 genotypes from 6 different populations of an ecotype of Achillea multifida, which is endemic to Uludag region of Turkey and classified among the plants that are at risk of becoming extinct, were characterized in terms of several parameters? of conservation biology and conservation genetics by using molecular markers.ITS and EST-SSRs, which are commonly used molecular marker types were used in this dissertation research. For the analysis of the results, Arlequin 3.11 and Phylip 3.67 software were used Ath the end of our research, values of Fst and standards heterozygisity were found to be 0.02984 and 0.4118, respectively, indicating a little genetic differentiation. In addition, the estimates of Nei's of gene diversity (Nm) parameters were 2.81.In conclusion, a total of 9 EST-SRR markers that could be used for population genetic studies in A. mulitifida. Our results did not show any signs of decrease in the genetic variation of Uludag populations of this plant.
Koruma Biyolojsi, türlerin, onların habitatlarının ve ekosistemlerinin yok olmasını önlemek amacıyla, yeryüzünün biyolojik çeşitliliğini ve doğasını inceleyen bilim dalıdır. Buna karşın koruma genetiği ise, koruma altındaki türlerin çevreye uyum dinamiklerini inceleyen genetik uygulamadır.Çalışmamızda Asteraceae familyasından Achilea multifida bitkisinin koruma genetiği ve biyolojisi çalışılmıştır. Uludağ endemiği olan ve yok olma riski kategorisine alınmış olan Achilea multifida bitkisinin 6 farklı popülâsyondan toplanan yaklaşık 50 genotipinin, farklı moleküler markör bölgesi çalışılmıştır.Çalışmamızda moleküler marker metotlarından ITS ve EST-SSR kullanılmıştır ve moleküler analizler sonucunda Fst ve ortalama heterozigotluk değerimiz sırasıyla 0.02948 ve 0.4118 olarak bulunmuştur. Aynı zamanda Nei'ye göre ortalama genetik çeşitlilik (Nm) 2.81 olarak bulunmuştur.Çalışmamız sonucunda türümüze ait 9 EST-SSR marker bulunmuştur. Yapılan genetik analizler sonucunda ise popülasyonlarda, genetik çeşitlilik açısından herhangi bir azalma gözlemlenmemiştir.
