Abstract

6. ÖZET Yaşlan 20-30 arasındaki sağlıklı kişilerden sağlanan kan örneklerinden elde edilen T lenfositlerinin kromozomları üzerinde yapılan bu çalışmada, hücre kültürü ve in-silu hibridizasyon yöntemlerinin optimizasyonu ve uygulanması ile, ürünü bağımsız bir aterosklerotik risk faktörü olan Lp(a) geninin yerleşim bölgesi, 6 'inci kromozomun uzun kolunun ucunda gösterilmiştir. Yapılan çalışma sırasında hücre süspansiyonunun preparata damlatıldıktan sonra geçen kuruma süresini ve preparat hazırlandıktan sonra geçen yaşlandmna zamanının, sonuçta alınan sinyalin kalitesini belirlemede önemli faktörler oldukları belirlenmiştir. In-silu hibridizasyon yönteminden verimli sonuç alabilmek için sağlanması gereken koşullar; iyi hazırlanmış ve yeterli sayıda metafaz plağı içeren bir preparat, bu preparatın en az bir hafta yaşlandınlması, digoksigenin ile en yüksek verimle işaretlenmiş 0. l-0.5pmol/15ul final konsantrasyonunda prob, 1:1000 dilüsyonda anti-digoksigenin alkalen fosfataz konjugatı ve probun baz dizisi içeriğine, uzunluğuna ve yıkama solüsyonunun [Na+] bağlı olarak belirlenen uygun hibridizasyon soması yıkama sıcaklığıdır. Bundan sonra yapılacak çalışmalarda serum Lp(a) düzeyi bilinen ve serum Lp(a) düzeyinin belirlenemediği durumlarda ft High resolution banding technique * ve in-siiu hibridizasyon yöntemleri ile Lp(a)'nın allellik formlarının belirlenebilmesinde, çalışmamızın temel teşkil edeceği düşüncesindeyiz. 46

7. SUMMARY By applying cell culture and in-situ hybridization techniques into T lymphocyte chromosomes obtained from healthy persons blood samples whose ages ranged between 20-30, we found that the Lp(a) gene whose product is an independent atherosclerotic risk factor, is localized at the end of the long arm of the sixth chromosome. During our work we found that, the drying time and age of the slide, in in-situ hybridization method are important factors to detect the signals in high quality. The necessary precautions to obtain good results from the in-situ hybridization method are; a well prepared slide with sufficient number of metaphase figures, the slide to be at least one week old, a probe at 0.1- 0.5pmole/l 5j.il final concentration which is labeled efficiently with digoxigenin, 1:1000 dilution of anti-digoxigenin alkalene phosphatase conjugate and a posthybridization wash temperature determined according to the probes base sequence, length and [Na+] concentration of the washing solution. In our future studies we are planning to detect the allelic forms of Lp(a) together with high resolution banding and in-situ hybridization techniques, in persons with detectable and undetectable serum Lp(a) concentrations. 47
