Abstract

ÖZET Bu çalışmada denenen Blok Haemat oxy lin-Eosin boyası histolojik boyama yöntemlerine literatürün ışığı altında de ğişik bir metod olarak sunulmuştur. Bu boyama yöntemiyle do- kular parafine gömülmeden önceki işlemlerle birlikte üstün bir şekilde boyanmaktadır. Bu boyalı bloklar aradan uzun yıl lar geçse bile bozulmamakta ve istenildiği anda boyalı ke sitlerin elde edilmesi mümkün olmaktadır. Blok Haematox lin-Eosin kesitlerinden elde edilen ke sitlere farklı amaçlar için değişik teknikleri uygulamak da mümkündür. Bunun için biz de laborat u var larda sıklıkla kullanılan, bağ dokularını boyayan Masson Trichorome Heiden- hain Azan (Azocarmine) boyalarını, dokulardaki glikojen ve glikozaminoglikanlar ı (GAG) boyayan PAS reaksiyonu metodu nu nukleusları ayırıcı bir şekilde boyayan Kernechtrot bo yalarını uyguladık. Alınan sonuçlar, literatür bilgileriyle karşılaştırıl mış, tartışılmış ve denenen metodun uygulanması tavsiye edil m i ş t i r.

SUMMARY The Block Haema t oxy 1 in-Eosin stain is used in diffe rent way to the histological methods in this study. The tissues are stained perfectly with this method, before embedding in paraffine. The stained blocks can be kept for a long time in a good state and they can be sectioned when needed. It is also possible to apply to the sections diffe rent technics. We used, therefore, Masson Trichrorae and, Heidenhain Azan (Azo-carmine), for staining connective tissues, PAS reactions for glycogen and gl icosaminogl yeans (GAG) in tissues and Kernechtrott dye for definitely staining nuclei. The results are compared with previous technics.
