Canine collagen type III glomerulopathy

Abstract

Collagen type III glomerulopathy (Col3GP) is a rare renal disease that is characterized by massive glomerular accumulations of collagen type III. The disease occurs in humans worldwide, typically with a sporadic occurrence. However, several cases in siblings have been observed, leading to the assumption that Col3GP is a hereditary disease. The exact cause and pathogenesis of this disease remain unknown. Marked serum concentration elevations of the aminoterminal propeptide of type III procollagen (PIIINP) is considered a useful marker for the disease in humans. In veterinary medicine, a few cases have been reported, including Col3GP in dogs, pigs, monkeys and cat. The background of the present study was the discovery of Col3GP in six related mixed-breed dogs. Further studies of the pedigree segregating Col3GP, including 12 additional Col3GPaffected offspring, indicated a genetic background with a simple autosomal recessive mode of inheritance.

Kollagen type III glomerulopati (Col3GP) er en sjelden nyresykdom som karakteriseres av uttalte glomerulære avleiringer av kollagen type III. Årsaken og patogenesen til sykdommen er ukjent, men den er antatt å være arvelig på grunn av økt forekomst i enkelte familier. Markert forhøyede serumnivåer av aminoterminal prekollagen III propeptid (PIIINP) har blitt funnet hos mennesker med Col3GP, og dette proteinet brukes derfor i humanmedisin som en markør for sykdommen. Hos dyr har sykdommen tidligere vært beskrevet hos hund, ape, gris og katt, i form av kasusrapporter. Bakgrunnen for denne studien var at Col3GP ble diagnostisert hos seks beslektede blandingshunder. Nye kull i denne hundefamilien ble fulgt, og i løpet av en femårsperiode ble ytterligere 12 affiserte avkom identifisert. Studiene av denne hundefamilien gav sterke holdepunkter for at Col3GP hos hund er en arvelig sykdom med autosomal recessiv arvegang.
