Abstract

La producción de ovocitos y la secreción hormonal en los ovarios, determinan la eficiencia reproductiva y en consecuencia, productiva de bovinos de leche y carne. La progresión a través del desarrollo folicular requiere de una comunicación entre el ovocito, las células de la granulosa y las células de la teca; y en dicho intercambio de información participan integrantes de la superfamilia del TGF-β (factor de de crecimiento transformante-β). El presente estudio fue diseñado para determinar la localización celular específica y expresión de los integrantes del sistema activina/inhibina/foliculostatina, de las isoformas del TGF-β y los receptores que podrían participar en la enfermedad quística ovárica en bovinos y en la persistencia folicular asociada. El desarrollo de los modelos experimentales resultó apropiado para el estudio de los estadios iniciales de la enfermedad así como el desarrollo de la persistencia encontrándose algunas diferencias entre los modelos y los casos espontáneos, las cuales podrían explicarse principalmente por el tiempo de persistencia. Se demostró que existe un posible efecto de activina, foliculostatina e inhibina como moduladores de la producción de hormonas esteroides, balance proliferación/apoptosis y diferenciación celular descriptas previamente en ovarios quísticos. Conociendo la fuerte justificación económica para desarrollar medidas preventivas y tratamientos para las alteraciones foliculares, los resultados obtenidos en esta tesis aumentan el conocimiento de la etiopatogenia de la enfermedad quística ovárica y los mecanismos involucrados en la persistencia folicular proveyendo una invaluable herramienta para estudios adiciones así como para la evaluación de medidas preventivas y terapéuticas.

The production of oocytes and ovarian hormone secretion, determine the reproductive efficiency and consequently productive efficiency in dairy and beef cattle. Progression through the follicular development requires a communication between the oocyte, granulosa cells and theca cells. In the exchange of information between these cells are involved members of the TGF-β superfamily (transforming growth factor-β). The present study was designed to determine the specific cellular localization and expression of the members of the activin/inhibin/folistatin system and isoforms of TGF-β and receptors who could participate in ovarian cystic disease in cattle and in the follicular persistence associated. The development of the experimental models was appropriate for the study of the early stages of the disease and the development of persistence found some differences between models and spontaneous cases, which could be explained mainly by the persistence time. We showed that there is a potential effect of activin, inhibin and folistatin as modulators of steroid hormone production, balance proliferation/apoptosis and cell differentiation previously described in cystic ovaries. Knowing the strong economic justification for developing preventive measures and treatments for follicular disorders, the results obtained in this thesis increase the knowledge of the pathogenesis of cystic ovarian disease and the mechanisms involved in the follicular persistence providing an invaluable tool for additional studies and for the evaluation of preventive and therapeutic actions.
