Abstract

Se ha realizado el presente estudio en vista de la polémica respecto al efecto del acetilsalicilato en el infarto de miocárdico. A perros mestizos de ambos sexos se les practicó ligadura de la arteria coronaria descendente anterior (3 horas) cuantificando el tamaño del infarto (nitroblue tetrazolium) como porcentaje en peso respecto al total del ventrículo izquierdo. Los perros se dividieron en un grupo control y cuatro grupos tratados con ASL, administrado por vía intravenosa, previamente a la ligadura coronaria y a las dosis de 30, 15, 7,5 y 3,5 mg/kg. El límite inferior de este rango de dosis fue elegido por ser un mínimo que aún inhibe, en esta especie animal, tanto la agregación plaquetaria inducida por ADP, como la vasodilatación coronaria inducida por ácido araquidónico, mientras la mayoría de estudios experimentales han utilizado dosis superiores a 10 mg/kg. Únicamente la dosis menor de ASL redujo de modo significativo el tamaño de la lesión isquémica. Esta misma dosis aumentó el flujo colateral retrógrado medido 1 hora postligadura, sin otras modificaciones significativas hemodinámicas o bioquímicas. En conclusión, los resultados de este estudio experimental muestran que sólo la dosis pequeña de ASL exhibe un efecto protector del micardio en este modelo experimental y plantea el interés de ensayar dosis sucesivamente menores.
