Abstract

RESUMEN: La polifenol oxidasa es la enzima responsable del pardeamiento de frutas y vegetales, en los que ocasiona disminución de las propiedades organolépticas y nutricionales. En este trabajo se evaluó la influencia de las condiciones de extracción (pH, concentración, composición del buffer y precipitación proteica) sobre la actividad de la polifenol oxidasa extraída de uchuva. La actividad polifenol oxidasa más alta se obtuvo al extraer la enzima utilizando buffer fosfato 0,2 M pH 6,0 suplementado con Triton X-100 y polivinil pirrolidona y concentrando las proteínas con acetona fría en una proporción sobrenadante:acetona 1:1. Posteriormente, la polifenol oxidasa se purificó parcialmente utilizando sistemas bifásicos acuosos formados por polietilenglicol y fosfato. Se evaluó el efecto de la composición de las fases, el peso molecular del polietilenglicol y el pH. Los mejores resultados se obtienen con un sistema formado por 5% de polietilenglicol 8000 y 14% de fosfato, en un pH de 5,5 con un factor de purificación de 4,65, un Ka de 4,70 y un porcentaje de rendimiento en la fase superior de 74,29%. Estos resultados indican un efecto significativo de las condiciones de extracción sobre la actividad polifenol oxidasa y proporcionan un método rápido, simple y eficiente para el aislamiento de esta enzima.

ABSTRACT: Polyphenol oxidase activity is responsible for the enzymatic browning of fruits and vegetables, and the end products of the catalyzed reaction are detrimental to food quality, in both sensory and nutritional properties. In this study we evaluated the influence of the extraction conditions (pH, concentration, composition of extraction buffer and protein precipitation agent) on the activity of golden berry polyphenol oxidase. The extraction of proteins with a mixture of TritonÆ X-100 and polyvinyl pyrrolidone in 0.2 M phosphate buffer at pH 6.0 and precipitation with cold acetone in a ratio supernatant-acetone of 1:1 gave the higher polyphenol oxidase activity. Partial purification of polyphenol oxidase was achieved using an aqueous two-phase system composed by polyethylene glycol and phosphate. Effect of phase composition, molecular weight of polythylene glycol and pH of the system on enzyme partitioning was studied. The optimum system was found at pH 5.5 containing 5% polythylene glycol 8000 and 14% phosphate, with Ka of 4.70, purification factor of 4.65 and a 74.29% yield of enzyme activity in the top phase. These results show a significant effect of extraction conditions on polyphenol oxidase activity and provide a quick, simple and efficient method for polyphenol oxidase isolation.
