Abstract

RESUMEN: Se evaluó el efecto del filtrado crudo del hongo Phytophthora infestans, agente causal del tizón tardío de la papa, y la respuesta de resistencia en callos en dos variedades de papa: Diacol Capiro (susceptible) y Diacol Monserrate (con resistencia de campo). También se estandarizaron en estas dos variedades los procesos de inducción, multiplicación de callos y regeneración de vitroplantas a partir de los mismos. Con la combinación hormonal 2,4D (1 mg/l) y BAP (2 mg/l) se obtuvo la mejor respuesta para inducir la formación de callos a partir de bojas jóvenes en los dos genotipos. La adición de tiosulfato de plata (1.0 mg/l) y ácido cítrico (50 mg/l) fue muy importante para la multiplicación de los callos de la variedad Diacol Capiro, tanto para incrementar el crecimiento como para evitar la muerte del tejido. El porcentaje de regeneración de plantas a partir de los callos fue superior en Diacol Monserrate. Sin embargo, ambas variedades mostraron buena capacidad regenerativa de los calles hasta los cuatro subcultivos evaluados. La dilución del filtrado crudo (concentrado veinte veces en rotoevaporador) que permitió la diferenciación de las variedades por la respuesta de resistencia a nivel de callos fue una parte del filtrado y cinco de medio de cultivo (1:5). Con este trabajo se propone un esquema de selección in vitro a partir de callos para ser usado en programas de mejoramiento para la resistencia a Phytophthora infestans.

ABSTRACT: The effect of the culture filtrate of Phytophthora infestans fungus, which is the causal agent of potato late bligth, and the resistance response at the callus level of two potatoes varieties, Diacol Capiro (susceptible) and Diacol Monserrate (field resistance), was evaluated. Whith the hormonal combination 2,4D (1 mg/l) and BAP (2 mg/l) was the best to induce callus formation of young leaves for the two genotypes. To the callus multiplication the addition of silver thiosulfate (1.0 mg/l) and citric acid (50 mg) was very important in Diacol Capiro to increase the growth and to avoid tissue death. The percent of the regeneration was higher in Diacol Monserrate but both varieties showed good regenerative capacity in callus with four subcultures. The dilution of the culture filtrate (twenty times rotoevapored) that best show the differential response resistance was 1:5. With this work we are presenting a scheme of in vitro selection to use in breeding programs to Phytophthora infestans resistance.
