Tratamos neste trabalho dos fundamentos e métodos técnicos da electroforese, cujo interesse, como processo de investigação, parece desnecessário encarecer. Recordam-se, primeiramente, algumas noções relacionadas com a carga eléctrica da molécula proteica e comportamento desta num campo eléctrico. Descreve-se, a traços largos, a técnica, digamos, clássica, de electroforese em célula de vidro, analisando, com os sistemas de leitura, as características da separação obtida, assim como o significado das curvas. Trata-se por último, das técnicas de electroforese em papel de filtro, que analisam com maior pormenor, apontando-se as suas possibilidades e limitações.
This paper deals with the fundamentals and technical methods of electrophoresis, the interest of which, as a research process, seems unnecessary to emphasise. Firstly, we recall some notions related to the electrical charge of the protein molecule and its behaviour in an electrical field. The technique of electrophoresis in a glass cell is described in broad terms, analysing the characteristics of the separation obtained, as well as the significance of the curves. Finally, the filter paper electrophoresis techniques are analysed in greater detail, pointing out their possibilities and limitations.
