Abstract

As benzodiazepinas são um grupo de fármacos cujos efeitos benéficos incluem a redução de ansiedade, indução e manutenção do sono, relaxamento muscular e tratamento e prevenção de ataques epiléticos. No entanto, existem alguns riscos associados à sua toma e são fármacos passíveis de abuso e uso indevido, pelo que são uma substância de interesse para a Toxicologia Forense. A Toxicologia Forense é a aplicação da toxicologia ao serviço da Lei. Envolve várias áreas: toxicologia forense post-mortem, ante-mortem comportamental e de desempenho humano, e análises para controlo de consumo de determinadas substâncias. Para realizar análises toxicológicas para a deteção e quantificação de benzodiazepinas podem ser usadas várias amostras biológicas. Como as matrizes biológicas são normalmente complexas, é necessário que as amostras passem primeiro por um processo de extração. Para a análise de benzodiazepinas podem ser utilizados diversos métodos analíticos, sendo os mais pertinentes os métodos cromatográficos e imunoensaios. Os imunoensaios são úteis para a deteção de benzodiazepinas, cuja presença numa amostra biológica é depois confirmada e quantificada por um método cromatográfico. Dentro dos métodos cromatográficos, destaca-se a cromatografia gasosa e a cromatografia líquida de alta eficiência. A cromatografia líquida de alta eficiência acoplada com espectrometria de massa em tandem é um dos métodos mais representados na literatura, uma vez que permite quantificar as benzodiazepinas com grande sensibilidade e especificidade.

Benzodiazepines are a class of drugs with several beneficial effects, including anxiolytic, hypnotic, muscle relaxant and anticonvulsant effect. However, there are some risks associated to its use and they can be object of abuse and misuse; therefore, they are frequently present in forensic cases. Forensic Toxicology is the use of toxicology at the service of the Law. It involves several areas: post-mortem forensic toxicology, behavioral and related to human performance ante-mortem forensic toxicology and drug testing. To detect and quantify benzodiazepines through toxicologic analyses, several biological samples can be used. Because biological matrices are generally very complex, it’s first necessary that they go through an extraction process. The analysis of benzodiazepines can be performed using several analytical methods. Among them, immunoassays and chromatographic methods are the most relevant. Immunoassays are useful to do an initial screening, followed by confirmation and quantification by a chromatographic method. Gas chromatography and high performance liquid chromatography are the most prominent within chromatographic methods. High performance liquid chromatography coupled with tandem mass spectrometry is one of the most described methods in scientific literature since it allows great sensibility and specificity in the quantification of benzodiazepines.
