Abstract

Staphylococcus aureus é uma das inúmeras bactérias que estão presentes no leite. É considerada uma das espécies mais patogénicas do género Staphylococcus, pois algumas das suas estirpes estão associadas a surtos de intoxicações alimentares. Estas estirpes produzem enterotoxinas, sendo algumas delas termoestáveis. Por forma a prevenir a contaminação do ser humano com as mesmas, o principal objetivo deste trabalho foi desenvolver um método de biologia molecular, mais especificamente um PCR multiplex, de modo a fazer simultaneamente a deteção dos genes das enterotoxinas estafilocócicas em amostras de leite cru e leite pasteurizado. Este é um método fácil, rápido, sensível e viável. As toxinas estafilocócicas são transmitidas maioritariamente por gado leiteiro, em animais infetados com mastites. As amostras (n=33) foram recolhidas de diferentes espécies de animais, grandes e pequenos ruminantes, incluindo caprinos (n=11), bovinos (n=7) e ovinos (n=5) e ainda foram recolhidas amostras de leite pasteurizado (n=10). Apenas 20 amostras de leite (60,6%), tiveram crescimento bacteriano para S. aureus: os bovinos tiveram 6, os caprinos 9 e os ovinos 5. Destas 20 estirpes apenas 4 (20%) apresentaram expressão do gene da enterotoxina SEC.O leite é um alimento muito presente na dieta da população portuguesa e cada vez mais devemos estar atentos a quaisquer possíveis contaminantes com impacto negativo na saúde pública, considerando que este trabalho poderá ser um contributo em prol da Saúde Pública. Ao diagnosticar animais infetados com mastite, como estes têm que ser tratados, é também preservada a saúde animal.

Staphylococcus aureus is one of the countless bacteria present in milk. It’s considered one of the most pathogenic species of the genus Staphylococcus, because some of the strains of this microorganism are often associated with food poisoning. These strains produce enterotoxins, and some of them are resistant to high temperatures. To help to prevent the human contamination with that toxins, the main objective of this study was the development of a molecular test, specifically, a multiplex PCR, to detect the staphylococcal enterotoxins genes in raw milk and pasteurized milk samples. This method is easy, sensitive, fast, and viable. Staphylococcal enterotoxins are mostly transmitted for dairy cattle, through animals infected with mastitis. The samples (n=33) were collected from different species, including goats (n=11), cows (n=7), and sheep (n=5) and also pasteurized milk (n=10). Only 20 samples showed bacterial growth for S. aureus: bovines had 6, goats 9 and sheep 5 positives. Only four of these 20 strains (20%) presented the expression of the SEC gene.Milk is a very important food in the diet of the Portuguese population, and we must pay attention to any possible contaminants with a negative impact on public health, considering that this work could be a contribution in favor of Public Health. When diagnosed with mastitis, these animals have to be treated and consequently the animal health is also preserved.
