Abstract

Introdução: O tratamento endodôntico visa eliminar a presença de microrganismos do interior dos canais e promover a reparação tecidual através do preenchimento tridimensional e selagem dos canais radiculares e acessórios. O material utilizado deve ser biocompatível para os tecidos peri-radiculares e ter a capacidade de formar uma barreira com a dentina canalar. Para promover a cicatrização e restauração da função dentária, os materiais dentários devem ser restauradores e biologicamente neutros.Objetivo: Avaliar os efeitos citotóxicos do cimento GuttaFlow Bioseal® e compará-los aos efeitos do cimento AH 26® na cultura celular MDPC-23. Materiais e Métodos: A linha celular MDPC-23 foi cultivada. Os meios de cultura foram condicionados durante 24h a 37ºC com os pellets de cimento GuttaFlow Bioseal® e AH 26®. As culturas celulares foram tratadas com o meio condicionado (1:1) e diluições seriadas (1:2, 1:4, 1:8 e 1:16). A fim de avaliar a atividade metabólica e viabilidade celular, foram realizados o ensaio MTT e SRB. Para determinar a produção de espécies reativas de oxigênio utilizaram-se as sondas de DHE e DCF-DA. O ciclo e a morte celular foram determinados por citometria, e para avaliar a capacidade de mineralização recorreu-se à coloração Alizarina red S. Resultados: A resposta das células expostas ao meio condicionado GuttaFlow Bioseal® demonstrou uma diminuição na atividade metabólica quando as células foram expostas às maiores concentrações (1:1 e 1:2), sendo mais evidente para períodos de incubação mais longos. No entanto, a manutenção da atividade metabólica é assegurada para concentrações mais baixas (1:4 a 1:16). De maneira semelhante, a viabilidade foi mantida após a incubação com o meio condicionado nas concentrações de 1:4 e 1:16, mas uma diminuição de 70% foi observada após a exposição à maior concentração. Observou-se um desequilíbrio na produção de peróxidos e anião superóxido. Quando a cultura celular foi exposta ao cimento AH 26® observou-se uma diminuição da atividade metabólica, contudo a viabilidade celular não foi comprometida. A produção de ROS foi alterada significativamente após o tratamento com o GuttaFlow® bioseal. Adicionalmente verificou-se ativação da morte por apoptose tardia e necrose e os resultados preliminares do ciclo celular evidenciam alterações.Conclusão: O cimento GuttaFlow Bioseal® apresentou alguma citotoxicidade dependendo do tempo e da concentração, o que pode estar relacionado à produção de ROS. O AH 26® demonstrou ser mais citotóxico pela maior morte celular e desregulação do ciclo, celular. Estes resultados encorajam estudos adicionais para entender a influência dos cimentos endodônticos no microambiente oral.

Introduction: Endodontic treatment aims to eliminate the presence of microorganisms inside the canals and promote tissue repair through three-dimensional filling and sealing of root and accessory canals. The material used must be biocompatible for the peri-radicular tissues and have the ability to form a barrier with the canalar dentin. To promote healing and restoration of dental function, dental materials must be restorative and biologically neutral.Objective: To evaluate the cytotoxic effects of GuttaFlow® Bioseal cement and to compare them to the effects of AH 26® cement in the MDPC-23 cell culture.Materials and Methods: The MDPC-23 cell line was cultured. The culture media were conditioned for 24h at 37 ° C with the GuttaFlow® Bioseal and AH 26® cement pellets. Cell cultures were treated with the conditioned medium (1: 1) and serial dilutions (1: 2, 1: 4, 1: 8 and 1:16). In order to evaluate metabolic activity and cellular viability, the MTT and SRB assays were performed. To determine the production of reactive oxygen species, the DHE and DCF-DA probes were used. Cycle and cell death were determined by cytometry, and to evaluate the mineralization capacity we resorted to Alizarin red S coloration.Results: The response of the cells exposed to the GuttaFlow Bioseal conditioned medium demonstrated a decrease in metabolic activity when cells were exposed to the highest concentrations (1: 1 and 1: 2), being more evident for longer incubation periods. However, maintaining metabolic activity is assured for lower concentrations (1: 4 to 1:16). Similarly, viability was maintained after incubation with the conditioned medium at concentrations of 1: 4 and 1:16, but a 70% decrease was observed after exposure to the highest concentration. An imbalance was observed in the production of peroxides and superoxide anion. When cell culture was exposed to AH 26® cement, a decrease in metabolic activity was observed, however, cell viability was not compromised. ROS production was significantly altered after treatment with GuttaFlow® bioseal. Additionally, activation of late apoptosis and necrosis death was observed and the preliminary results of the cell cycle showed changes.Conclusion: GuttaFlow® Bioseal cement showed some cytotoxicity depending on time and concentration, which may be related to ROS production. AH 26® has been shown to be more cytotoxic by increased cell death and cell cycle dysregulation. These results encourage further studies to understand the influence of endodontic cements in the oral microenvironment.
