Abstract

O acetato de 16-desidropregnenolona (16-DPA) é um esteroide que apresenta um esqueleto muito versátil, que permitiu a síntese de diferentes fármacos esteróides, tais como dexametasona, betametasona e inibidores da 5α-redutase. Existem poucos exemplos na literatura científica da reação de Diels-Alder envolvendo este composto como dieno ou como dienófilo, utilizando irradiação de micro-ondas (MO). Com o objetivo de sintetizar novos derivados do acetato de 16-desidropregnenolona utilizaram-se 1,3-diarilpropenonas e o 1- acetilciclopenteno como modelo de reatividade e foram exploradas as suas capacidades, como dieno ou como dienófilo, na reação de Diels-Alder sob condições de reação diferentes. A influência do solvente e dos catalisadores foram estudados sob aquecimento convencional e sob irradiação de micro-ondas. Após a realização do estudo das condições de reação utilizouse o esteroide 16-DPA como dienófilo e o ciclopentadieno, ou o 2,3-dimetil-1,3-butadieno como dienos. As suas estruturas dos produtos obtidos foram elucidadas por análise comparativa com o reagente utilizando ressonância magnética nuclear de protão e espectrometria de massa.

16-Dehidropregnenolona Acetate (16-DPA) has a very versatile skeleton which has allowed the synthesis of different steroidal drugs such as dexamethasone, β-metasona and inhibitors of 5α-reductase. There are a few examples in the scientific literature of the Diels- Alder reaction involving this compound as diene or dienophile and using microwave irradiation (MO). Aiming to synthesize new dehydropregnenolone derivatives we used 1,3- diarylpropenones and 1-acetylciclopentene as template to explored reactivity their capacity as diene or dienophile in the Diels-Alder reaction under different reaction conditions. The influence of different solvents and catalysts were studied under conventional heating and microwave irradiation. Once explored the reaction conditions we used the steroid 16-DPA as dienophile and cyclopentadiene or 2,3-dimethyl-1,3-butadiene as dienes. The structures of the reaction products were elucidated by comparative analysis with the reagent using proton nuclear magnetic resonance and mass spectrometry.
