Abstract

O microbioma assume hoje um papel central na saúde humana, animal e vegetal, sendo considerado um alvo estratégico no âmbito da perspetiva One Health. O avanço da engenharia do microbioma tem permitido o desenvolvimento de abordagens inovadoras com impacto direto na promoção da saúde, na sustentabilidade agrícola e no bem-estar animal. Entre as principais estratégias encontram-se o uso de prebióticos e probióticos, o recurso a bacteriófagos naturais e sintéticos, os consórcios microbianos, o transplante de microbiota fecal e vaginal e as ferramentas de manipulação genética. Contudo, a complexidade das interações microbianas, a competição com microbiotas nativas e a ausência de métodos eficazes de monitorização in situ permanecem continuam a limitar a aplicação destas abordagens em larga escala. Além disso, a dificuldade em manipular geneticamente microrganismos de interesse representa outro entrave, exigindo a identificação e caracterização de novos plasmídeos conjugativos, o desenvolvimento de métodos alternativos de transferência de DNA e uma compreensão mais profunda dos sistemas de defesa microbianos. Perspetiva-se que os avanços futuros se centrem em tratamentos personalizados, apoiados por tecnologias de sequenciação de nova geração, pela integração de dados genómicos, metagenómicos e metabólicos, e pelo uso de algoritmos de aprendizagem automática. Deste modo, a presente monografia descreve as principais estratégias de engenharia do microbioma, discutindo os seus impactos, limitações e perspetivas futuras da área enquanto alternativa promissora às terapêuticas convencionais.

The microbiome currently plays a central role in human, animal, and plant health, being considered a strategic target within the One Health perspective. Advances in microbiome engineering have enabled the development of innovative approaches with a direct impact on health promotion, agricultural sustainability, and animal welfare. Among the main strategies are the use of prebiotics and probiotics, the application of natural and synthetic bacteriophages, microbial consortia, fecal and vaginal microbiota transplantation, and genetic manipulation tools. However, the complexity of microbial interactions, competition with native microbiotas, and the lack of effective in situ monitoring methods continue to limit the large-scale application of these approaches. In addition, the difficulty in genetically manipulating microorganisms of interest represents another barrier, requiring the identification and characterization of new conjugative plasmids, the development of alternative methods for DNA transfer, and a deeper understanding of microbial defense systems. Future advances are expected to focus on personalized treatments, supported by next-generation sequencing technologies, the integration of genomic, metagenomic, and metabolic data, and the use of machine learning algorithms. Thus, the present monograph describes the main strategies of microbiome engineering, discussing their impacts, limitations, and future perspectives in the field as a promising alternative to conventional therapies.
