Effects of N,N-Dimethyltryptamine (DMT) on the Functional Connectivity of the Third Visual Pathway

Abstract

N,N-Dimethyltryptamine (DMT) is a classical psychedelic that induces profound and immersive altered states of consciousness. This substance is characterized by its interaction with serotonin 5-HT2A receptors, leading to changes in abilities such as perception and empathy. However, the mechanisms by which DMT alters brain connectivity, particularly in regions associated with social perception, remain poorly understood. The third visual pathway (TVP) is a critical neural circuit involved in the processing of biological motion and social signals, playing a key role in interpreting facial expressions, following gaze direction, and processing other cues essential for social cognition and theory of mind (ToM).Previous studies indicate that psychedelics such as Lysergic Acid Diethylamide (LSD) and psilocybin enhance neuroplasticity and alter functional connectivity in regions associated with sensory, emotional, and cognitive processing. These studies provide evidence that classical psychedelics, including DMT, induce increased brain activity and connectivity, associated with altered states of perception. This study aims to explore the effects of DMT on the functional connectivity of the TVP, considering its potential implications for social cognition, including perception, empathy, and ToM. Based on previous research findings, it is hypothesized that DMT increases functional connectivity between TVP regions and areas associated with emotional processing and social cognition. To test this hypothesis, a within-subject experimental design was employed, consisting of an active condition (DMT-induced state) and a control condition, separated by a minimum interval of one month. In the active condition, participants inhaled 50-70 mg of DMT extract derived from the root bark of Mimosa hostilis, with a concentration of 30.92%. In the control condition, no substance was administered. Eleven healthy adults with prior experience in DMT use were recruited. Data acquisition was conducted using resting-state functional magnetic resonance imaging (rs-fMRI) immediately after DMT inhalation. Functional connectivity analysis was performed using a seed-based approach, with seeds defined in TVP regions to assess connectivity changes between TVP regions and other brain areas. Additionally, a region-of-interest (ROI)-to-ROI connectivity analysis was conducted between regions within the TVP to investigate intrinsic connectivity changes within the pathway induced by DMT.The results revealed increased connectivity between TVP regions during the active condition, indicating enhanced processing of visual information through this pathway. Furthermore, data indicated increased functional connectivity between the TVP and regions of the default mode network (DMN) and the limbic system, which are involved in the integration of socio-emotional information. These changes suggest that DMT may alter the integration of visual and emotional signals, enhancing the capacity to interpret complex social stimuli, such as facial expressions and gaze direction. In conclusion, DMT has a significant impact on the functional connectivity of the TVP, suggesting changes in both visual and emotional processing. These findings imply that DMT may influence social and emotional abilities, particularly in contexts of social interaction, by modulating critical neural networks involved in social cognition. These results support further research into the potential therapeutic applications of DMT for neuropsychological conditions characterized by deficits in social cognition and interaction, such as Autism Spectrum Disorder (ASD) and social anxiety.
