Abstract

O presente relatório descreve as competências adquiridas e as atividades desenvolvidas ao longo do estágio curricular, no âmbito do segundo ano do Mestrado em Genética Clínica e Laboratorial da Faculdade de Medicina da Universidade de Coimbra, com duração de nove meses. O estágio decorreu entre setembro de 2023 e maio de 2024 e teve lugar na Unidade Funcional de Hematologia Molecular- Sector de Hemato-Oncologia, localizado no Hospital Pediátrico da Unidade Local de Saúde de Coimbra.O estágio teve como objetivo o acompanhamento da rotina laboratorial do setor de Hemato-Oncologia com foco no estudo das alterações genéticas essenciais para o diagnóstico, classificação e estratificação de risco da leucemia mieloide aguda. Durante o decorrer do estágio curricular, tive a possibilidade de executar diferentes técnicas de biologia molecular, bem como participar na implementação de novos protocolos para pesquisa de mutações nos genes KMT2A e UBTF.Durante o estágio curricular estive em contacto com diferentes tipos de amostras biológicas (sangue periférico, medula óssea e raiz de cabelo), bem como diversas técnicas laboratoriais, nomeadamente, extração de ácidos nucleicos, PCR convencional, eletroforese em gel e eletroforese capilar, PCR em tempo real, sequenciação de Sanger e Next Generation Sequencing.Este relatório contém uma breve descrição do laboratório, as diferentes técnicas de biologia molecular executadas durante o estágio, a caracterização molecular e a otimização de dois novos estudos genéticos num grupo de doentes com leucemia mieloide aguda.

This report describes the skills acquired during the internship, as well as the activities carried out during the second year of the master’s degree in Clinical Laboratory Genetics at the Faculty of Medicine of the University of Coimbra, which lasted nine months. The internship took place between September 2023 and May 2024 at the Molecular Haematology Functional Unit - Haemato-Oncology Sector, located at the Paediatric Hospital of the Coimbra Local Health Unit.The aim of the internship was to integrate the laboratory routine of the Haematology-Oncology sector, focusing on the study of genetic alterations essential for the diagnosis, classification, and risk stratification of acute myeloid leukaemia. In the course of the internship, I was able to perform different molecular biology techniques and participate in the implementation of novel protocols for researching mutations in the KMT2A and UBTF genes.During my internship I encountered different types of biological samples (peripheral blood, bone marrow and hair roots), as well as various laboratory techniques, namely nucleic acid extraction, conventional PCR, gel electrophoresis and capillary electrophoresis, real-time PCR, Sanger sequencing and Next Generation Sequencing.This report contains a brief description of the laboratory, the different molecular biology techniques carried out during the internship, the molecular characterisation and optimisation of two new genetic studies on a group of patients with acute myeloid leukaemia.
