Abstract

A biomonitorização humana (HBM - do Inglês Human Biomonitoring) tem-se revelado uma ferramenta eficaz na avaliação da exposição individual, nomeadamente a micotoxinas. A micotoxina Zearalenona (ZEA) apresenta principalmente propriedades estrogénicas atuando como desregulador endócrino no sistema reprodutor, apresentando também um perfil imunotóxico, hematotóxico, hepatotóxico e genotóxico, aspetos que sustentam a necessidade de monitorizar a sua exposição. Assim, o estudo de HBM realizado na presente dissertação tem como objetivos determinar os níveis de ZEA em amostras de urina através do imunoensaio enzimático ELISA(do Inglês Enzyme Linked Immunosorbent Assay), avaliar o risco de exposição à ZEA por parte das crianças com idade entre os cinco e os 12 anos e identificar os fatores antropométricos, sociodemográficos, caraterísticas individuais e os hábitos alimentares determinantes para a exposição das crianças. Para isso, foram recolhidas 42 amostras de urina de crianças, entre os cinco e os 12 anos, do sexo masculino e feminino residentes no distrito de Santarém, e analisadas pelo imunoensaio enzimático ELISA, após extração em fase sólida (SPE).O presente estudo demonstrou que cerca de 76% das amostras analisadas encontravam-se contaminadas por ZEA, com concentração média de 188,12±253,99 ng/mL. Para a avaliação de exposição, foi calculada a ingestão diária provável (PDI) e comparada com a ingestão diária tolerável (TDI) estabelecida pela EFSA com um valor de 0,25 µg/kg. O quociente de perigo variou entre 0,01 e 4,01, apontando a ZEA como uma preocupação para a Saúde Pública. Apenas o gelado apresentou uma correlação estatisticamente significativa com o teor de ZEA. Assim, são necessários estudos adicionais para obter uma avaliação de risco mais robusta, considerando as caraterísticas inerentes às diferentes populações, os efeitos tóxicos desta micotoxina e a frequência de deteção determinada.

Human Biomonitoring (HBM) reveals to be an efficient tool for individual exposure assessment, for example of mycotoxins. The mycotoxin Zearalenone (ZEA) exposure needsmonitoring due to the fact that besides being known as an estrogenic mycotoxin acting as an endocrine disruptor at reproductive system, it also shows an imunotoxic, hematoxic,hepatotoxic and genotoxic profile.Thus, the HBM study performed in this thesis aims to determinate ZEA levels in urinesamples through Enzyme-Linked Immunosorbent Assay ELISA, assess ZEA exposure risk tochildren aged between five and 12 years old and identify anthropometric and sociodemographicfactors, individual characteristics and eating habits that can be determinant for children´s exposure.For this purpose, 42 samples were collected from girls and boys aged between five and 12 years old and residing in district of Santarém, and analysed by ELISA enzyme immunoassayafter solid-phase extraction (SPE).The present study showed that about 76% of the samples were contaminated with ZEA, with a mean concentration of 188,12±235,99 ng/mL. Futhermore, for the exposure assessement, the probable daily intake (PDI) was calculated and compared with tolerable daily intake (TDI) established by EFSA with a value of 0,25 µg/kg. Hazard Quotient varied between 0,01 and 4,01 showing that ZEA is a concern for Public Health. Only ice-cream showed a statistically significant correlation with ZEA level.Therefore, further studies are needed to obtain a more robust risk assessment, considering different population´s characteristics, the toxic effects of this mycotoxin and the given detection´s frequency.
