Abstract

Un virus recombinant dépourvu du gène sf32 (Sf32null), unique au nucléopolyhédrovirus multiple de Spodoptera frugiperda (SfMNPV), a été généré par recombinaison homologue à partir d'un bacmide comprenant le génome viral complet (Sfbac). L'analyse transcriptionnelle a révélé que sf32 est un gène précoce. Les corps d'occlusion (OBs) de Sf32null contenaient 62 % plus d'ADN génomique que les virus contenant le gène sf32, Sfbac et Sf32null-repair, bien que l'ADN de Sf32null soit trois fois moins infectieux lorsqu'il est injecté in vivo. Les OBs Sf32null avaient un diamètre 18 % plus grand et contenaient 17 % plus de nucléocapsides dans les ODV que ceux de Sfbac. Aucune différence significative n'a été détectée dans la pathogénicité de l'OB (concentration létale de 50%), la vitesse de destruction ou la production de virus bourgeonné in vivo. En revanche, la production d'OBs/larves a été réduite de 39% chez les insectes infectés par Sf32null par rapport à ceux infectés par Sfbac. Nous concluons que SF32 est une protéine non essentielle qui pourrait être impliquée dans l'organisation des nucléocapsides pendant l'assemblage ET l'occlusion des ODV, entraînant une augmentation du nombre de nucléocapsides dans les ODV.

Se generó un virus recombinante que carecía del gen sf32 (Sf32null), exclusivo del nucleopoliedrovirus múltiple de Spodoptera frugiperda (SfMNPV), mediante recombinación homóloga a partir de un bácmido que comprende el genoma viral completo (Sfbac). El análisis transcripcional reveló que sf32 es un gen temprano. Los cuerpos de oclusión (OB) de Sf32null contenían un 62% más de ADN genómico que los virus que contenían el gen sf32, Sfbac y Sf32null-repair, aunque el ADN de Sf32null fue tres veces menos infeccioso cuando se inyectó in vivo. Las OB Sf32null eran un 18% más grandes en diámetro y contenían un 17% más de nucleocápsides dentro de las ODV que las de Sfbac. A recombinant virus lacking the sf32 gene (Sf32null), unique to the Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV), was generated by homologous recombination from a bacmid comprising the complete viral genome (Sfbac). Transcriptional analysis revealed that sf32 is an early gene. Occlusion bodies (OBs) of Sf32null contained 62% more genomic DNA than viruses containing the sf32 gene, Sfbac and Sf32null-repair, although Sf32null DNA was three-fold less infective when injected in vivo. Sf32null OBs were 18% larger in diameter and contained 17% more nucleocapsids within ODVs than those of Sfbac. No significant differences were detected in OB pathogenicity (50% lethal concentration), speed-of-kill or budded virus production in vivo. In contrast, the production of OBs/larva was reduced by 39% in insects infected by Sf32null compared to those infected by Sfbac. The SF32 predicted protein sequence showed homology (25% identity, 44% similarity) to two adhesion proteins from Streptococcus pyogenes and a single N-mirystoylation site was predicted. We conclude that SF32 is a non-essential protein that could be involved in nucleocapsid organization during ODV assembly and occlusion, resulting in increased numbers of nucleocapsids within ODVs.
