Molecular methods for detecting guar additions to locust bean gum.

Abstract

[EN]The invention concerns methods for detecting guar gum alone or in mixtures of guar gum and locust bean gum. The invention also concerns methods for extracting, amplifying and detecting DNA of guar and locust bean gums and the mixtures thereof. DNAs of plants from which guar and locust bean gums are extracted are amplified by means of polymerase chain reaction (PCR) using conserved initiators. Differences in the sequences of the amplification products obtained from these two plants enable their differentiation, the identification of guar DNA in mixtures of guar and locust bean mixtures and the design of guar-specific PCR initiators which detect guar only in mixtures of guar and locust bean gums. [ES]Esta invención describe los métods para detectar la goma de guar con goma de garrofín. Describe los métodos para la extracción, amplificación y detección del DNA de las gomas de guar y garrofín y de sus mezclas. El DNA de las plantas de las que se extraen las gomas de guar y garrofín se amplifican mediante la cadena en reacción de la polimerasa PCR usando unos iniciadores conservados. Las diferencias en las secuentcias de los productos de amplificaciçon obtenidos a partir de esas dos plantas permiten su diferenciación, la identificación de DNA de guar en mezclas de gomas de guar y garrofín, y el diseño de iniciadores de la PCR específicos del guar que lo detectan sólo en mezclas de gomas de eguar y garrofín.
