UJI AKTIVITAS ANTIOKSIDAN EKSTRAK KULIT BUAH NAGA (Hylocereus Monacanthus)

Abstract

Kulit buah naga bisa dijadikan sebagai obat herbal alami yang dimanfaatkan sebagai antioksidan. Antioksidan merupakan senyawa yang mampu menghambat ataupun menangkal proses oksidasi lipid. Sebagian besar penyakit tubuh dipicu oleh radikal bebas yang merupakan bahan yang berbahaya. Pada umumnya pemanfaatan buah naga merah hanya pada bagian daging buahnya saja, sehingga kulit buah naga yang dihasilkan masih berupa limbah yang belum dimanfaatkan potensinya. Keberadaan antioksidan pada kulit buah naga perlu diidentikasi dan dioptimalkan pemanfaatannya. Untuk melihat potensi antioksidan pada kulit buah naga, maka perlu dilakukan studi lebih lanjut. Tujuan penelitian ini yaitu untuk mengetahui keberadaan antioksidan dan nilai IC50 ekstrak kulit buah naga. Metode penelitian yang digunakan adalah eksperimental, menggunakan metode DPPH (1,1-difenil-2-pikrilhidrazil) dengan seri konsentrasi ekstrak yang dibuat yaitu 2; 4; 6; 8 dan 10 ppm. Pengukuran aktivitas antioksidan ekstrak kulit buah naga ditentukan sampai diperoleh IC50 dengan memasukan nilai y (y=50) pada persamaan garis y = bx + a. Absorban yang dihasilkan diukur menggunakan alat Spektrofotometer UV-Vis pada panjang gelombang 515 nm. Berdasarkan hasil penelitian didapatkan ekstrak kulit buah naga mengandung aktivitas antioksidan dengan kadar IC50 sebesar 9,16 setelah dihitung menggunakan persamaan garis regresi y = 0,0054x + 0,5061. sementara IC50 asam askorbat yag digunakan sebagai standar diperoleh nilai 1,47. Nilai IC50 kulit buah naga dinyatakan memiliki kandungan aktivitas antioksidan yang sangat kuat. Aktivitas antioksidan fraksi etanol kulit buah naga diduga karena adanya senyawa metabolit sekunder golongan alkaloid, flavonoid, tanin dan saponin.
