A Virus-Derived Stacked RNAi Construct Confers Robust Resistance to Cassava Brown Streak Disease

Abstract

La enfermedad de la raya marrón de la yuca (CBSD, por sus siglas en inglés) amenaza la seguridad alimentaria y económica de los pequeños agricultores de África Oriental y Central, y representa una amenaza para la producción de yuca en África Occidental. El CBSD es causado por dos especies de virus transmitidos por la mosca blanca: el virus de la raya marrón de la yuca (CBSV) y el virus de la raya marrón de la yuca ugandesa (UCBSV) (género: Ipomovirus, familia Potyviridae). Aunque se han logrado diferentes niveles de tolerancia a través de la cría convencional, hasta la fecha, no se ha identificado una resistencia efectiva a CBSD dentro del germoplasma de yuca de África Oriental. La tecnología de ARNi se utilizó para integrar la resistencia a CBSD en el cultivar de yuca preferido por los agricultores ugandeses TME 204. Las líneas de plantas transgénicas que acumulan niveles bajos, medios y altos de ARNip se expusieron a injertos de yemas con el aislado virulento de CBSV Naliendele solo o en combinación con UCBSV. La resistencia a CBSD en el invernadero se correlacionó directamente con los niveles de ARNip derivados de CP según lo determinado por la evaluación visual de los síntomas de las hojas y las raíces de almacenamiento, y el diagnóstico por RT-PCR de la presencia de los patógenos. Se encontró que las líneas de baja expresión eran susceptibles a CBSV y UCBSV, mientras que las líneas de plantas de acumulación media a alta eran resistentes a ambas especies de virus. La ausencia de virus detectable en las líneas transgénicas p5001 de mejor rendimiento se confirmó adicionalmente mediante inoculación inversa a través de exposición a savia o injerto a Nicotiana benthamiana y cultivar de yuca 60444 susceptibles a CBSD, respectivamente. Los datos presentados muestran una resistencia robusta de las líneas transgénicas p5001 TME 204 tanto a CBSV como a UCBSV en condiciones de invernadero. La maladie des stries brunes du manioc (CBSD) menace la sécurité alimentaire et économique des petits agriculteurs d'Afrique de l'Est et d'Afrique centrale et constitue une menace pour la production de manioc en Afrique de l'Ouest. La CBSD est causée par deux espèces de virus transmis par les aleurodes : le virus de la traînée brune du manioc (CBSV) et le virus de la traînée brune du manioc ougandais (UCBSV) (genre : Ipomovirus, famille des Potyviridae). Bien que différents niveaux de tolérance aient été atteints grâce à la sélection conventionnelle, à ce jour, la résistance efficace au CBSD dans le germoplasme du manioc d'Afrique de l'Est n'a pas été identifiée. La technologie RNAi a été utilisée pour intégrer la résistance au CBSD dans le cultivar de manioc TME 204 préféré des agriculteurs ougandais. Des lignées végétales transgéniques ont été générées exprimant une construction répétée inversée (p5001) dérivée de séquences de protéines de revêtement (CP) de CBSV et UCBSV fusionnées en tandem. Des Northern blots utilisant des sondes spécifiques pour chaque séquence de CP ont été effectués pour caractériser 169 lignées transgéniques indépendantes pour l'accumulation de siRNA dérivés de CP. Des lignées de plantes transgéniques accumulant des niveaux faibles, moyens et élevés de siRNA ont été soumises à une greffe de bourgeon avec l'isolat virulent de Naliendele CBSV seul ou en combinaison avec l'UCBSV. La résistance à la CBSD dans la serre est directement corrélée aux niveaux de siRNA dérivés de la CP tels que déterminés par l'évaluation visuelle des symptômes foliaires et racinaires de stockage, et le diagnostic RT-PCR de la présence des agents pathogènes. Les lignées à faible expression se sont révélées sensibles au CBSV et à l'UCBSV, tandis que les lignées de plantes à accumulation moyenne à élevée étaient résistantes aux deux espèces de virus. L'absence de virus détectable dans les lignées transgéniques p5001 les plus performantes a été confirmée par rétro-inoculation via la sève ou la greffe au Nicotiana benthamiana sensible au CBSD et au cultivar de manioc 60444, respectivement. Les données présentées montrent une résistance robuste des lignées transgéniques p5001 TME 204 au CBSV et à l'UCBSV dans des conditions de serre. Les niveaux de résistance étaient directement corrélés aux niveaux d'expression de l'ARNsi dérivé du transgène, de sorte que ce dernier peut être utilisé comme prédicteur de la résistance à la CBSD.

Cassava brown streak disease (CBSD) threatens food and economic security for smallholder farmers throughout East and Central Africa, and poses a threat to cassava production in West Africa. CBSD is caused by two whitefly-transmitted virus species: Cassava brown streak virus (CBSV) and Ugandan cassava brown streak virus (UCBSV) (Genus: Ipomovirus, Family Potyviridae). Although varying levels of tolerance have been achieved through conventional breeding, to date, effective resistance to CBSD within East African cassava germplasm has not been identified. RNAi technology was utilized to integrate CBSD resistance into the Ugandan farmer-preferred cassava cultivar TME 204. Transgenic plant lines were generated expressing an inverted repeat construct (p5001) derived from coat-protein (CP) sequences of CBSV and UCBSV fused in tandem. Northern blots using probes specific for each CP sequence were performed to characterize 169 independent transgenic lines for accumulation of CP-derived siRNAs. Transgenic plant lines accumulating low, medium and high levels of siRNAs were bud graft challenged with the virulent CBSV Naliendele isolate alone or in combination with UCBSV. Resistance to CBSD in the greenhouse directly correlated to levels of CP-derived siRNAs as determined by visual assessment of leaf and storage root symptoms, and RT-PCR diagnosis for presence of the pathogens. Low expressing lines were found to be susceptible to CBSV and UCBSV, while medium to high accumulating plant lines were resistant to both virus species. Absence of detectable virus in the best performing p5001 transgenic lines was further confirmed by back-inoculation via sap or graft challenge to CBSD susceptible Nicotiana benthamiana and cassava cultivar 60444, respectively. Data presented shows robust resistance of transgenic p5001 TME 204 lines to both CBSV and UCBSV under greenhouse conditions. Levels of resistance correlated directly with levels of transgene derived siRNA expression such that the latter can be used as predictor of resistance to CBSD.
