Whole Genome-Based Amplified Fragment Length Polymorphism Analysis Reveals Genetic Diversity in Candida africana

Abstract

Cette étude visait à étudier la diversité génétique d'un panel de souches de Candida africana récupérées à partir d'échantillons vaginaux dans différents pays. Toutes les souches fongiques étaient hétérozygotes au locus de type accouplement et appartenaient au génotype A de Candida albicans. De plus, toutes les souches de C. africana examinées manquent d'assimilation de N-acétylglucosamine et l'analyse de séquence du gène HXK1 a montré un polymorphisme distinctif qui altère l'utilisation de ce sucre aminé dans cette levure. Le séquençage multi-locus de sept gènes domestiques a révélé une homogénéité génétique substantielle entre les souches, à l'exception des locus CaMPIb, SYA1 et VPS13 qui ont contribué de manière significative à la classification de notre ensemble de souches de C. africana en six types de séquences diploïdes existants. L'analyse des empreintes digitales du polymorphisme de la longueur des fragments amplifiés a donné une plus grande hétérogénéité génotypique parmi les souches de C. africana. Dans l'ensemble, les données rapportées ici montrent que la diversité génétique de C. africana existe et que l'existence de ce groupe intrigant de souches de C. albicans avec des phénotypes spécifiques associés pourrait être utile pour de futures études comparatives afin de mieux comprendre la génétique et l'évolution de cet important agent pathogène humain.

Este estudio tuvo como objetivo investigar la diversidad genética de un panel de cepas de Candida africana recuperadas de muestras vaginales en diferentes países. Todas las cepas fúngicas eran heterocigotas en el locus tipo apareamiento y pertenecían al genotipo A de Candida albicans. Además, todas las cepas de C. africana examinadas carecen de asimilación de N-acetilglucosamina y el análisis de secuencia del gen HXK1 mostró un polimorfismo distintivo que perjudica la utilización de este aminoazúcar en esta levadura. La secuenciación de múltiples locus de siete genes constitutivos reveló una homogeneidad genética sustancial entre las cepas, excepto para los loci CaMPIb, SYA1 y VPS13, que contribuyeron significativamente a la clasificación de nuestro conjunto de cepas de C. africana en seis tipos de secuencias diploides existentes. El análisis de huellas dactilares de polimorfismo de longitud de fragmento amplificado produjo una mayor heterogeneidad genotípica entre las cepas de C. africana. En general, los datos informados aquí muestran que en C. africana se produce diversidad genética y la existencia de este intrigante grupo de cepas de C. albicans con fenotipos específicos asociados podría ser útil para futuros estudios comparativos con el fin de comprender mejor la genética y la evolución de este importante patógeno humano.

This study aimed at investigating the genetic diversity of a panel of Candida africana strains recovered from vaginal samples in different countries. All fungal strains were heterozygous at the mating-type-like locus and belonged to the genotype A of Candida albicans. Moreover, all examined C. africana strains lack N-acetylglucosamine assimilation and sequence analysis of the HXK1 gene showed a distinctive polymorphism that impair the utilization of this amino sugar in this yeast. Multi-locus sequencing of seven housekeeping genes revealed a substantial genetic homogeneity among the strains, except for the CaMPIb, SYA1 and VPS13 loci which contributed significantly to the classification of our set of C. africana strains into six existing diploid sequence types. Amplified fragment length polymorphism fingerprint analysis yielded greater genotypic heterogeneity among the C. africana strains. Overall the data reported here show that in C. africana genetic diversity occurs and the existence of this intriguing group of C. albicans strains with specific phenotypes associated could be useful for future comparative studies in order to better understand the genetics and evolution of this important human pathogen.
