FISSURAS LABIAIS E/OU PALATINAS NÃO SINDRÔMICAS: DETERMINANTES AMBIENTAIS E GENÉTICOS.

Abstract

As fissuras labiais e/ou palatinas não sindrômicas (FL/PNS) são anomalias congênticas resultantes de defeitos na fusão dos processos craniofaciais. Estas alterações apresentam incidência variada e são mais comuns na forma não sindrômica. O objetivo deste trabalho foi realizar uma revisão da literatura sobre as FL/PNS, enfatizando os aspectos genéticos. Na análise crítica dos artigos selecionados observou-se que vários genes e regiões cromossômicas têm sido associados às fendas orofaciais em estudos de associação de larga escala genômica (GWAS), como os genes IRF6 , ABCA4 , MafB e região 8q24. Trata-se de um distúrbio de desenvolvimento de origem multifatorial, envolvendo agentes ambientais e genéticos. Os autores também observaram uma forte interferência de fatores étnicos, particularmente em estudos do tipo caso-controle. Alguns polimorfismos foram identificados em estudos de GWAS replicados e associados às fissuras na população brasileira, como o polimorfismo rs987525 , rs1530300 e rs560426. Quando a população é estratificada , como no Brasil, a composição étnica pode ter um forte efeito sobre a contribuição genética das FL/PNS. Recomenda-se o estudo da ascendência genética ssociado à investigação de polimorfismos genéticos sobre fissuras orofaciais.
