Characterization of the PlARG locus mediating resistance against Plasmopara halstedii in sunflower

Abstract

The resistance locus PlARG originates from the wild species Helianthus argophyllus and effectively protects sunflower (H. annuus) against all known races of downy mildew (Plasmopara halstedii). To determine the molecular structure of the PlARG locus fine mapping was performed in the population (cms)HA342 x ARG1575-2 encompassing 2145 F2 individuals. Cosegregating resistance gene candidates RGC151, RGC52a, and RGC52b as well as closely flanking markers were genetically mapped. Resistance gene candidates were isolated in a BAC library and additional candidate genes were identified by transcript analyses. Der Resistenzlocus PlARG stammt aus der Wildart Helianthus argophyllus und schützt die Kultursonnenblume H. annuus effektiv gegen alle bekannten Rassen des Falschen Mehltaus (Plasmopara halstedii). Um die molekulare Struktur des PlARG Locus aufzuklären, wurde eine Feinkartierung in der Population (cms)HA342 x ARG1575-2 mit 2145 F2 Individuen durchgeführt. Cosegregierende Resistenzgenkandidaten RGC151, RGC52 und RGC52b sowie eng gekoppelte Marker konnten genetisch kartiert werden. Resistenzgenkandidaten wurden aus einer BAC-Bibliothek isoliert und weitere Kandidatengene durch Transkriptanalysen identifiziert.
