PENGARUH EKSTRAK DAUN SIRSAK TERHADAP EKSPRESI GEN BCL2 SEL KANKER PAYUDARA T47D

Abstract

Kanker payudara merupakan jenis kanker dengan angka kejadian menempati posisi pertama terbanyak di Indonesia dibandingkan dengan jenis kanker lain. Daun sirsak memiliki kandungan acetogenin, memiliki efek sitotoksik terhadap sel kanker payudara T47D. Tujuan penelitian ini untuk mengetahui pengaruh ekstrak daun sirsak terhadap ekspresi gen BCL2 sel kanker T47D secara in vitro. Penelitian ini merupakan penelitian eksperimental, menggunakan metode relative kuantifikasi real time PCR menggunakan cell line T47D diinkubasi selama 72jam yang dibagi menjadi 2 kelompok sampel, yaitu kontrol dan perlakuan. Kelompok perlakuan diberi larutan uji ekstrak etanol daun sirsak dengan konsentrasi sesuai IC50 = 94,26 μg/ml. Hasil PCR akan memunculkan nilai Cq, yang mana akan dianalisis dengan menggunakan uji man whitney dan rumus livak. Hasil penelitian menunjukkan sel kanker payudara yang diberi perlakuan mengekspresikan gen BCL2 yaitu 1,85 kali lipat lebih tinggi dibandingkan dengan sel kelompok kontrol. Terdapat perbedaan yang bermakna antara ekspresi gen BCL2 sel kanker payudara T47D kelompok perlakuan dan kontrol dengan nilai p = 0,001 (p < 0,005).Ekspresi gen BCL2 pada sel kanker payudara T47D yang diberi perlakuan ekstrak daun sirsak (Annona Muricata Linn) lebih tinggi dari pada kontrol.
