Abstract

Daun sungkai merupakan salah satu tanaman obat Indonesia yang sering digunakan sebagai obat pilek, obat cacing, pencegah sakit gigi dengan cara berkumur, campuran rempah di air mandi bagi wanita yang baru saja melahirkan dan sebagai penurun panas. Penelitian ini dilakukan untuk mengetahui kandungan senyawa metabolit sekunder pada ekstrak daun sungkai serta menentukan aktivitas anti bakteri, toksisitas dan inhibitor enzim tirosinase. Ekstraksi daun sungkai di lakukan dengan cara maserasi dengan menggunakan pelarut butanol yang telah di variasikan konsentrasi pelarutnya yaitu 95%, 75%, dan 50%. Ekstrak daun sungkai yang telah didapatkan di uji kandungan metabolit sekunder nya, didapatkan bahwa ekstrak butanol daun sungkai mengandung fenolik, steroid dan alkaloid. Pada uji antibakteri dengan menggunakan bakteri Salmonella Thypi yaitu semua ekstrak bersifat kuat kecuali esktrak butanol 75% dan 50% pada konsentrasi 5% dan 2,5% di dapatkan zona inhibisi sebesar 9 mm. pada bakteri Lysteria monocytogenets semua ekstrak memiliki aktivitas anti bakteri yang kuat. Hasil dari uji toksisitas dengan menggunakan metode Brine Shrimp Lethality Test (BSLT) terlihat bahwa ekstrak butanol 95%, 75%, dan 50% bersifat toksik yaitu dengan nilai LC50 dari ekstrak tersebut adalah 49.2153 mg/L, 97.8152 mg/L, dan 495.450 mg/L. Pada uji inhibitor enzim tyrosinase didapat nilai IC50 dari ekstrak butanol 95%, 75% dan 50% sebesar 2,839.88 mg/L, 2,213.66 mg/L, dan 3,963,23 mg/L dan dapat dikatakan bahwa ekstrak butanol daun sungkai tidak memiliki kekuatan penghambatan.
