Abstract

Enzimi su biološki katalizatori. Oni ubrzavaju kemijske reakcije ne mijenjajući konstantu ravnoteže već smanjujući energiju aktivacije. Po svom sastavu su proteini, s tim što postoje i katalitički aktivne ribonukleinske kiseline koje se nazivaju ribozimi. Imenuju se prema specifičnim pravilima koja jasno opisuju reakcije koje određeni enzimi kataliziraju. Izrazito su specifični što znači da svaki enzim katalizira točno određenu reakciju, odnosno ima aktivno mjesto prilagođeno za određeni supstrat. Aktivno mjesto je ono u kojem se odvija pretvorba supstrata u produkt preko nastanka enzim – supstrat kompleksa. Enzimski katalizirane reakcije opisuju se Michaelis – Menten modelom iz kojeg se izvodi izraz koji povezuje početnu brzinu reakcije s koncentracijom supstrata te se iz njega mogu dobiti sve informacije o promatranoj reakciji. Isto tako postoje različiti mehanizmi enzimski kataliziranih reakcija kao i načini regulacije enzimske aktivnosti. Specifičnost enzima dovedena je u pitanje nakon što je definiran pojam koji se naziva promiskuitet enzima. To je svojstvo enzima da katalizira dodatne reakcije pored glavne reakcije koju katalizira. Ove dodatne reakcije su uglavnom mnogo sporije od glavne reakcije koju katalizira promiskuitetni enzim. Iako su dodatne reakcije fiziološki irelevantne, promiskuitet enzima s evolucijskog stajališta može se reći da služi kao početna točka za razvoj novih funkcija enzima kroz mutacije ili usmjerenu laboratorijsku evoluciju. Mehanistički aspekt nastoji objasniti zašto dolazi do promiskuiteta enzima. Zaključak je da promiskuitetne aktivnosti enzima nisu loše već su vrlo korisne te, između ostalog, pomažu razvoju biotehnoloških sustava.
