Phenotypic Analysis of Circulating Myeloid Derived Suppressor Cells and Their Subpopulations in Egyptian Females with Breast Cancer: A Single-Centre Case-Control Study

Abstract

Les cellules suppressives dérivées de la myéloïde (MDSC) sont des cellules myéloïdes immatures avec une fonction suppressive qui a été soigneusement documentée dans le contexte du cancer. Notre objectif était d'évaluer les niveaux de MDSC et de leurs sous-ensembles dans une cohorte de patientes égyptiennes atteintes d'un cancer du sein. L'évaluation de la MDSC totale dans le sang périphérique et de son sous-ensemble a été effectuée à l'aide de la cytométrie en flux multicolore chez 30 patientes malignes, 10 patientes bénignes atteintes d'une tumeur du sein et 10 patientes témoins en bonne santé. Les patientes atteintes de CB présentaient des niveaux de MDSC totale plus élevés par rapport aux témoins (p= 0,01), en particulier la MDSC monocytaire (M-MDSC) et les sous-ensembles de MDSC anormaux (p = 0,001 et p 2 cm présentait des MDSC granulocytaires (G-MDSC) significativement plus élevées par rapport à une taille de tumeur < 2 cm (p= 0,02) alors que les MDSC anormales étaient significativement plus élevées chez les patientes avec une taille de tumeur < 2 cm (p = 0,037). MDSC et ses sous-ensembles peuvent être utilisés comme marqueur pronostique de la taille de la tumeur ainsi que comme cibles potentielles pour le traitement chez les patientes atteintes de cancer du sein.

Las células supresoras derivadas de mieloides (MDSC) son células mieloides inmaduras con función supresora que se ha documentado a fondo en el contexto del cáncer. Nuestro propósito fue evaluar los niveles de MDSC y sus subconjuntos en una cohorte de pacientes egipcios con cáncer de mama. La evaluación de MDSC totales en sangre periférica y su subconjunto se realizó mediante citometría de flujo multicolor en 30 pacientes con tumores de mama malignos, 10 pacientes con tumores benignos y 10 mujeres de control sanas. Las pacientes con CMB tenían niveles de MDSC totales más altos en comparación con los controles (p= 0.01), particularmente los subconjuntos de MDSC monocíticos (M-MDSC) y MDSC anormales (p = 0.001 y p 2 cm mostró MDSC granulocíticas (G-MDSC) significativamente más altas en comparación con un tamaño tumoral < 2 cm (p= 0,02), mientras que las MDSC anormales fueron significativamente más altas en pacientes con un tamaño tumoral < 2 cm (p = 0,037). Las MDSC y sus subconjuntos se pueden utilizar como marcador pronóstico del tamaño tumoral, así como objetivos potenciales para el tratamiento en pacientes con cáncer de mama.

Myeloid-derived suppressor cells (MDSC) are immature myeloid cells with suppressive function that has been thoroughly documented in the setting of cancer. Our purpose was to evaluate levels of MDSC and their subsets in a cohort of Egyptian patients with breast cancer.Evaluation of peripheral blood total MDSC and its subset was done using multicolor flowcytometry in 30 malignant, 10 benign breast tumor patients and 10 healthy control females.BC patients had higher total MDSC levels compared to controls (p= 0.01) particularly the Monocytic MDSC (M-MDSC) and abnormal MDSC subsets (p = 0.001 and p 2 cm exhibited significantly higher granulocytic MDSCs (G-MDSCs) compared to tumor size < 2 cm (p= 0.02) whereas abnormal MDSCs were significantly higher in patients with a tumor size < 2 cm (p = 0.037).MDSC and its subsets can be used as a prognostic marker of tumor size as well as a potential targets for treatment in breast cancer patients.

الخلايا الكابتة المشتقة من النخاع (MDSC) هي خلايا نخاعية غير ناضجة ذات وظيفة قمعية تم توثيقها بدقة في بيئة السرطان. كان هدفنا هو تقييم مستويات MDSC ومجموعاتها الفرعية في مجموعة من المرضى المصريين المصابين بسرطان الثدي. تم إجراء تقييم لمجموع الدم المحيطي MDSC ومجموعته الفرعية باستخدام قياس التدفق متعدد الألوان في 30 مريضًا خبيثًا و 10 مرضى أورام ثدي حميدة و 10 إناث مراقبة صحية. كان لدى مرضى BC مستويات MDSC إجمالية أعلى مقارنة بعناصر التحكم (p= 0.01) خاصةً مجموعات MDSC أحادية الخلية (M - MDSC) ومجموعات MDSC الفرعية غير الطبيعية (p = 0.001 و p 2 سم مستويات أعلى بكثير من MDSCs المحببة (G - MDSCs) مقارنة بحجم الورم < 2 سم (p= 0.02) في حين أن MDSCs غير الطبيعية كانت أعلى بكثير في المرضى الذين يعانون من حجم الورم < 2 سم (p = 0.037). يمكن استخدام MDSC ومجموعاته الفرعية كعلامة تنبؤية لحجم الورم بالإضافة إلى أهداف محتملة للعلاج في مرضى سرطان الثدي.
