Abstract

Цель исследования – анализ изменений липидной компоненты мембран эритроцитов экспериментальных животных при действии различных стрессорных факторов и возможность коррекции их с помощью водно-спиртового экстракта из туники асцидии пурпурной. Серия экспериментов проводилась на крысах линии Вистар с массой тела 180-200 г. Экстракт из туники асцидии пурпурной получали методом реперколяции на 40% этиловом спирте при соотношении сырья к экстрагенту 1:1 (по объему). Сухой остаток составляет 6-7%. Экстракт вводили внутрижелудочно в дозе 0,4 мл/кг массы животного, что соответствует 28 мг/кг сухого остатка. В экстрактах общих липидов эритроцитов с помощью хроматографических методов определяли количественное содержание фосфолипидов. В условиях экспериментального стресса в мембранах эритроцитов отмечалось изменение соотношения отдельных фосфолипидных фракций. При этом нарушения липидной компоненты затрагивало как внешний, так и внутренний монослой мембраны: снижалась концентрация основных структурных фосфолипидов с одновременным увеличением количества их лизоформ. Такие перестройки в липидной составляющей способствуют ухудшению эластических свойств мембраны. Применение экстракта из туники асцидии пурпурной позволяет улучшить биохимические показатели, максимально приблизить их к контрольным значениям. Таким образом, экстракт обладает восстановительным действием и может быть рекомендован в качестве эффективного средства для профилактики стресса. The purpose of the study – analysis of changes of the lipid components of erythrocyte membranes in experimental animals under the impact of different stress factors and the possibility of correcting it by using hydroalcoholic extract from the purple ascidian’s tunic. Experiment was performed on Wistar rats with body weight of 180-200 g. The extract from the purple ascidian’s tunic were obtained by the method of repercolation with 40% ethyl alcohol. The yield was 1 liter extract from 1 kg of raw material. The dry residue is 6-7%. The extract was administered intragastrically at a dose of 0.4 ml/kg of animal weight, which corresponds to 28 mg/kg dry residue. In extracts of total lipids of erythrocytes by chromatographic methods was determined the quantitative content of phospholipids. Under the impact of stress factors changes in the ratio of individual phospholipid fractions were observed. At the same time, disturbances of the lipid component affected both the outer and inner monolayer of the membrane: the contents of the major phospholipids of the membranes decreased with a simultaneous increase in the amount of lysoforms. Such rearrangements in the lipid component contribute to the deterioration of the elastic properties of the membrane. The use of extract from the purple ascidian’s tunic allows to bring biochemical parameters to control values. Thus, the extract has a restorative effect and can be recommended as an effective remedy for preventing stress.
