Abstract

Цель работы: коррекция физиолого-биохимических характеристик эритроцитов крови крыс при гиперхолестериновом рационе экстрактом из морской бурой водоросли Saccharina japonica. Исследование проводили на крысах линии Вистар массой 180-200 г, содержавшихся в стандартных условиях вивария. Животные были разделены на 3 группы по 10 крыс в каждой: 1-я - контроль (интактные); 2-я группа - гиперхолестериновый рацион; 3-я группа - гиперхолестериновый рацион + экстракт сахарины. Гиперхолестеринемию вызывали в течение 6 недель путем введения в рацион повышенного содержания насыщенных жиров (растительное сало 25% от веса рациона), включающих 2,5% холестерина. Экстракт из водоросли вводили перорально в виде водного раствора сухого остатка (предварительно освобожденный от спирта экстракт путем упаривания в вакууме) в дозе 100 мг/кг массы общих полифенолов в сутки. Физиологические характеристики эритроцитов (объем, диаметр, осмотическая резистентность) определяли по общепринятым методам. Фракционное разделение фосфолипидов из мембран эритроцитов осуществляли методом двумерной микротонкослойной хроматографии. Гиперхолестериновый рацион способствовал увеличению объема эритроцитов, сдвигу осмотической резистентности, увеличению в мембране количества холестерина, сфингомелина, лизофракций фосфолипидов, а также снижению фосфатидилинозита и фосфатидной кислоты. Введение экстракта из Saccharina japonica способствовало нормализации изученных параметров эритроцитов. Objective: correction of physiological and biochemical characteristics of red blood cells of rats with hypercholesterolemic ration by the extract from sea brown alga Saccharina japonica. The study was carried out on Wistar rats weighing 180-200 g, maintained in standard vivarium conditions. The animals were divided into 3 groups of 10 rats in each: 1st - control (intact); group 2 - hypercholesterolemic diet; group 3 - hypercholesterolemic diet + extract from Saccharina japonica. Hypercholesterolemia was caused for 6 weeks by introducing a high content of saturated fats (vegetable fat of 25% of the weight of the ration), including 2,5% of cholesterol, into the diet. Extract from the algae was administered orally once a day in the form of an aqueous solution of a dry residue (previously freed from alcohol extract by evaporation in a vacuum) at a dose of 100 mg / kg of total polyphenols. Physiological characteristics of erythrocytes (volume, diameter, osmotic resistance) were determined by conventional methods. Fractional separation of phospholipids from erythrocyte membranes was performed by two-dimensional micro thin-layer chromatography. Hypercholesterolemic diet promoted increase the erythrocyte’s volume, shift in osmotic resistance, increase the amount of cholesterol, sphingomyelin, lysophospholipids, as well as decrease the amount of phosphatidylinositol and phosphatidic acid. The administration of Saccharina japonica extract promoted the normalization of the studied parameters of red blood cells.
