Цель исследования - изучить влияние модельных взвесей микроразмерных твердых взвешенных частиц (ТВЧ) на морфофункциональное состояние и пероксидацию липидов культуры клеток альвеолярных макрофагов (АМ). Из бронхоальвеолярного лаважа (БАЛ) 17 крыс линии Вистар выделяли и культивировали АМ. Макрофаги нагружали модельными взвесями ТВЧ, идентичных по составу и содержанию частиц атмосферного воздуха районов с незначительной (взвесь № 1) и высокой (взвесь № 2) техногенной нагрузкой. Морфофункциональное состояние культуры клеток макрофагов до- и после воздействия взвесей ТВЧ изучали визуально с помощью светового микроскопа. Ответную реакцию оценивали по динамике гидроперекисей липидов (ГПЛ) и общего антиоксидантного статуса (TAS) в культуре клеток и среде их культивирования. Установлено, что культура клеток АМ наиболее активно фагоцитирует микрочастицы модельной взвеси № 2, в большей степени обогащенной кварцевыми фракциями РМ1, чем частицы модельной взвеси № 1. В результате такого воздействия значительно повышается содержание гидроперекисей липидов в АМ. Сохранение ТАS при воздействии обеих модельных взвесей на культуру АМ на уровне интактной группы культуры макрофагов свидетельствует о неспособности АМ с незавершенным фагоцитозом поддерживать высокий потенциал антиоксидантный защиты, сдерживающий образование активных форм кислорода (АФК) и переокисление липидов мембран. Полученные результаты свидетельствуют, что модельная взвесь, идентичная по составу и содержанию мелкодисперсных ТВЧ атмосферному воздуху района с высокой техногенной нагрузкой, вызывает наиболее выраженную окислительную модификацию липидов и способствует развитию «окислительного взрыва» в культуре АМ. Интенсивность оказанного воздействия модельных взвесей зависит как от содержания в них легко фагоцитируемых фракций ТВЧ - PM1 и PM2,5, а также и от содержания в них частиц кварцевой природы, усиливающих продукцию свободных радикалов в клетках, вызывающих морфофункциональные изменения макрофагов и гибель клеток. The aim. To study the influence of model suspensions of micro-sized suspended particulate matter (SPM) on the morphofunctional condition and peroxidation of lipids in cell cultures of alveolar macrophages (AM). Materials and methods. AM were isolated from bronchoalveolar lavage (BAL) of 17 Vistar's line rats and cultivated. Macrophages were loaded by model suspensions of SPM, which were identical in the composition and the content of particles of atmospheric air of areas with insignificant (suspension № 1) and high (suspension № 2) technogenic load. Morphofunctional condition of cell culture of macrophages before and after the impact of suspensions of SPM was studied visually by light microscope. The response was evaluated by dynamics of lipid hydroperoxides (HPL) and total antioxidant status (TAS) in the cell culture and their culture medium. Results. It is found, that cell culture of АМ more actively phagocytose microparticles of model suspension № 2, that the most enriched by quartz fractions of РМ1, than the model suspension № 1. As a result of this impact, the content of lipid hydroperoxides in АМ is increased significantly. Preservation of ТАS during impact of both model suspensions on culture of АМ on the level of intact group of macrophage culture shows of inability of АМ with incompleted phagocytosis to support a high potential anti-oxidant protection, constraining formation of the active form of oxygen (AFO) and peroxidation of membrane lipids. Conclusions. Findings testify to model suspension, that is identical in the composition and the content of fine SPM of atmospheric air of the area with high technogenic load, causes the most expressed oxidative modification of lipids and promotes the development of «oxidative explosion» in АМ culture. The intensity of the impact of model suspensions depends on the presence of easy phagocytosed fractions of SPM - PM1 and PM2,5 and the quartz particles in them. These particles amplify the production of free radicals in cells, that cause morphological and functional changes of macrophages and cell death.
