Súhrn Malígne gliómy sú jedným z typov nádorov mozgu, ktoré najviac vzdorujú liečbe. Rozvoj rezistencie na rádioterapiu a chemoterapiu rovnako prispieva k agresívnym fenotypom týchto nádorov. Za posledných 50 rokov sa zistila zvýšená hladina lipidov a produktov ich metabolizmu v gliómoch. Molekulárne mechanizmy, vďaka ktorým neoplasticky transformované tkanivá získavajú a využívajú tieto lipidy, však doposiaľ nie sú dobre známe. V našej štúdii sme pozorovali 3 skupiny lipidov. V skupine fosfatidylcholínov sa počas skorého štádia rakoviny mozgu zistilo 48% signifikantne zmenených metabolitov u samíc a 66,2% metabolitov u samcov potkanov kmeňa Sprague Dawley. V skupine lyzofosfatidylcholínov bolo množstvo zmenených metabolitov 57,1% u samíc a 64,3% u samcov. Najzaujímavejšie výsledky sme pozorovali v skupine sfingomyelínov, kde vyše 85% metabolitov bolo signifikantne zvýšených pri rakovine mozgu oproti kontrolnej skupine u obidvoch pohlaví. Abstract Malignant gliomas are one of the most treatment-resistant types of brain tumors. The development of resistance to radiotherapy and chemotherapy also contributes to the aggressive phenotypes of these tumors. Over the past 50 years, an increased level of lipids and products of their metabolism has been found in gliomas. However, the molecular mechanisms by which neoplastically transformed tissues acquire and use these lipids are still not well understood. In our study, we observed 3 groups of lipids. In the phosphatidylcholine group, 48% of significantly altered metabolites in females and 66.2% of metabolites in male Sprague Dawley rats were found during the early stage of brain cancer. In the group of lysophosphatidylcholines, the amount of changed metabolites was 57.1% in females and 64.3% in males. We observed the most interesting results in the sphingomyelin group, where over 85% of metabolites were significantly increased in brain cancer compared to the control group in both sexes.
